皮肤血管瘤组织中CHFR和PIK1的表达及意义

摘要 目的探讨CHFR和PIK1在人皮肤血管瘤组织中的表达及其意义。方法收集武汉大学人民医院病理科2008年-2011年皮肤毛细血管瘤存档蜡块40例,其中男性15例,女性25例。采用免疫组织化学S-P法检测40例皮肤血管瘤增生期、退化期及正常皮肤组织CHFR和PIK1表达水平,采用图像分析系统对CHFR和PIK1的表达进行定量分析,并用SPSS13.0软件对各组免疫组织化学反应阳性颗粒的平均光密度、阳性面积率做单因素方差分析和SNK(q)检验。结果 (1)CHFR的表达增生期血管瘤血管内皮细胞中可见少量的棕黄色颗粒,CHFR呈低表达,正常皮肤组及退化组血管内皮细胞中可见密集分布的棕黄色颗粒,CHFR呈高表达。增生期组CHFR的表达明显低于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义(P>0.05)。(2)PIK1的表达增生期血管瘤血管内皮细胞中可见密集分布的棕黄色颗粒,PIK1呈高表达,正常皮肤组及退化组血管内皮细胞中可见少量的棕黄色颗粒,PIK1呈低表达。增生期组PIK1的表达明显高于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义(P>0.05)。结论 CHFR低表达促进了细胞周期,增强了细胞生长能力,与血管瘤的发生发展密切相关;在血管瘤增生期中,PIKl的表达水平上调,过量表达的PIK1会引起细胞的增殖,表明PIK1蛋白在血管瘤增生期中高水平表达与细胞的增殖是密切相关的。 Objective To study the role of CHFR and PIK1in human dermal hemangioma.Methods Specimens from 40cases(15males,and 25females)of hemangioma were collected from the Department of Pathology in Renmin Hospital of Wuhan University from 2008to 2011.We examined the expression of CHFR and PIK1in proliferative,involuting hemangiomas and normal skin tissues by using immunohistochemistry.Average optical density and positive area rates of the expression of CHFR and PIK1were measured by image analysis(HPIAS-1000).Results 1.CHFR expression in the endothelial cells was significantly lower in proliferative hemangioma than in involuting hemangioma and normal skin(P<0.05).No significant difference was found in the expression of CHFR between involuting hemangioma and normal skin(P >0.05).2.PIK1expression in the endothelial cells was significantly higher in proliferative hemangioma than in involuting hemangioma and normal skin(P<0.05).No significant difference was found in the expression of PIK1between involuting hemangioma and normal skin(P>0.05).Conclusion Low expression of CHFR promotes the cell cycle,enhancing the ability of cell growth that is closely related to the development of hemangioma.The excessive expression of PIK1protein in proliferative hemangioma is closely related to cell proliferation.
