肝刺激因子在MCD饮食喂养小鼠非酒精性脂肪性肝炎中的作用 被引量:1

摘要 目的探讨肝刺激因子(hepatic stimulator substance,HSS)对蛋氨酸胆碱缺乏饲料(methionine and choline-deficient diet,MCD)饮食引起的小鼠非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)的保护作用。方法采用MCD饮食进行小鼠NASH模型的制备,股静脉注射腺病毒包装HSS表达载体pAdxsi-Flag-HSS,用HE染色观察肝细胞组织改变,油红O染色观察肝内脂质沉积,并检测β-羟脂酰CoA脱氢酶(β-hydroxyacyl-COA dehydrongenase,HAD)的活性。结果 HSS的表达能显著减少MCD引起NASH小鼠肝细胞的脂滴的形成,并提高HAD的活性。结论 HSS对MCD造成的NASH肝损伤起到保护作用,这种保护作用可能部分通过提高HAD的活性而实现的。 Objective To investigate the protective effects of hepatic stimulator substance(HSS)on nonalcoholic steatohepatitis(NASH)induced by methionine and choline-deficient diet(MCD).Methods Mice were fed with an MCD diet.After four weeks liver weight,body weight,liver histological changes,activity ofβ-hydroxyacyl-COA dehydrongenase(HAD)were measured.Results NASH was remarkably alleviated in mice subjected to the HSS gene therapy.HSS increased the avtivity of HAD.Conclusion HSS protects hepatocyte from NASH in mice fed with an MCD diet.The protection provided by HSS may be partially via preservation of HAD.
