石蜡包埋细胞芯片的手工制备方法及其应用价值 被引量:2

摘要 目的介绍一种石蜡包埋细胞芯片的简易制备方法及验证其应用价值。方法收集科研培养细胞或临床细胞学标本,手工法制作石蜡包埋的细胞芯片,并进行HE染色、免疫组织化学法及TUNEL检测法验证细胞芯片的应用价值。结果此法熟练掌握后能制备出60个点阵左右的细胞芯片,肉眼观察点阵完整,排列均一,经HE染色、免疫组化染色及TUNEL凋亡染色证实,染色效果良好,结果可靠。结论简易手工法制备的石蜡包埋细胞芯片的操作容易,设备简单,能够迅速成型制备出多种不同类型的细胞芯片,值得推广应用。 Objective To introduce a simple preparation method of paraffin-embedded cell microarray and verify its application value.Methods Collecting cultured cells or clinical cytological specimens,we manuallu made paraffin-embedded cell microarray and verified its application value by HE staining,immunohistochemical staining and TUNEL technique.Results After mastering this method,we could produce cell microarray of about 60 lattices that were complete and uniform.The dyeing effect of the cell microarray was confirmed reliable by HE,immunohistochemistry and TUNEL.Conclusion As the manual preparation method of paraffin-embedded cell microarray is simple and easy to operate,the method can be conveniently used for cell microarray and is worth popularization and application.
