Wnt-1、Fzd-1在环磷酰胺致大鼠神经管畸形发生中的表达变化

摘要 目的观察Wnt-1、Fzd-1在环磷酰胺致大鼠神经管畸形发生中的表达变化,探讨神经管畸形的发生机制。方法将孕鼠随机分为实验组和对照组,孕13d8:00AM按15mg/kg体重分别腹腔注射环磷酰胺和生理盐水。注射后8、12、24、48h取胎鼠,制备石蜡连续切片。用免疫荧光技术检测Wnt-1、Fzd-1的表达变化。结果注射后8、12h,实验组Wnt-1、Fzd-1的表达与对照组相比无显著性差异;24、48h,Wnt-1、Fzd-1的表达强度均比对照组低,差异有统计学意义。结论环磷酰胺抑制Wnt-1、Fzd-1的表达,干扰Wnt经典通路的传导,是环磷酰胺致神经管畸形的原因之一。 Objective To explore the mechanism of neural tube defects(NTDs)induced by cyclophosphamide in rats by detecting the expression of Wnt-1and Fzd-1.Methods Pregnant rats were randomly divided into experimental and control groups.At 8:00AM on the 13 th day of pregnancy,the experimental groups were intraperitoneally injected with cyclophosphamide(15mg/kg body weight),while the control groups were injected with the same dose of normal saline.The pregnant rats were sacrificed at 8,12,24,48 hours after injection,then the fetuses were prepared into serial paraffin sections.The expressions of Wnt-1and Fzd-1were detected by immunofluorescence staining.Results Compared with those of the control groups,the expressions of Wnt-1and Fzd-1in experimental groups were not altered at 8,12 hours after injection.But the optical density of Wnt-1and Fzd-1in experimental groups declined significantly at24,48 hours after injection.Conclusion The conduction of the classic Wnt pathway is interfered by downregulation of Wnt-1and Fzd-1,which may be one of the important reasons for NTDs induced by cyclophosphamide.
