摘要 目的观察凋亡抑制因子Livin和磷酸化p38MAPK(p-p38MAPK)在乳腺癌组织中的表达,探讨其表达的临床意义。方法随机选取70例乳腺癌和对照组的30例癌旁组织蜡块标本,应用免疫组织化学技术检测Livin和p-p38MAPK蛋白的表达及细胞内定位,分析其表达与乳腺癌患者临床病理相关指标之间的关系。收集手术切除的20例新鲜乳腺癌和其相对应的癌旁组织标本,运用免疫印迹(Western blot)技术进一步定量检测Livin和p-p38MAPK蛋白的表达水平。并应用统计学软件Spearman等级相关检验分析Livin和p-p38MAPK蛋白表达的相关性。结果免疫组织化学结果显示,Livin、p-p38MAPK分别在64.3%(45/70)和57.1%(40/70)的乳腺癌组织中阳性表达,其阳性率均显著高于对照组的癌旁组织(P<0.05)。并且Livin、p-p38MAPK蛋白表达均与乳腺癌患者的不同TNM分期、有或无腋窝淋巴结转移密切相关(P<0.05)。Western blot亦进一步证实,Livin、p-p38MAPK蛋白在乳腺癌中的表达水平亦均高于其相对应的癌旁组织(P<0.05)。此外,Livin与p-p38MAPK蛋白表达呈正向相关性(r=0.318,P=0.008)。结论 Livin、p-p38MAPK蛋白的表达在乳腺癌的发展演进过程中可能起着不可忽视的作用,并且两者可能存在一定的调控关系。 Objective To investigate the expression of apoptosis inhibitor protein(Livin)and phosphorylated p38mitogen-activated protein kinase(p-p38MAPK)in breast cancer,and to explore their clinical significance.Methods Histological specimens of 70 cases of breast cancer tissues and 30 cases of paracancerous tissues(controls)were randomly selected.The expression and location of Livin and p-p38 MAPK proteins examined by immunohistochemical staining.The relationship of Livin and p-p38 MAPK with clinicopathological features of patients with breast cancer was analysed.Western blot was used to determine the protein expression levels of Livin and p-p38 MAPK in another 20 breast cancer samples and 20 adjacent tissues.The correlation of Livin and p-p38 MAPK was analysed by Spearman test.Results Immunohistochemistry showed that the positive expression rates of Livin and p-p38 MAPK in breast cancer were 64.3%and 57.1%,respectively,which were significantly higher than those in controls(P<0.05).The expression of both proteins was associated with TNM staging and lymph node metastasis(P<0.05 for all).Western blot also showed that the expression levels of Livin and p-p38 MAPK proteins in breast cancer tissues were higher than those in adjecent tissues(P<0.05 for all).Furthermore,the correlation analysis revealed that the expression of Livin and p-p38 MAPK proteins displayed a positive correlation(r=0.318,P=0.008).Conclusion Livin and p-p38 MAPK proteins may play an important role in the carcinogenesis and development of breast cancer,and they may act synergistically.
