电针降低氯胺酮大鼠岛叶皮质和尾壳核c-Fos水平 被引量:1

摘要 目的为了证明电针是否对氯胺酮滥用大鼠岛叶皮质(insular cortex,IC)和尾壳核(caudate putamen,CP)c-Fos表达产生影响。方法将32只清洁级SD大鼠采用随机数字表法,分为正常组、生理盐水组、模型组(氯胺酮腹腔注射)和电针组(氯胺酮+电针一侧"三阴交"和"足三里"穴)。采用免疫组织化学方法检测IC和CP内c-Fos表达。结果与正常组、生理盐水组相比,模型组大鼠出现鼠尾僵直上翘、扭体等兴奋症状;IC和CP内c-Fos阳性细胞数明显增多;与模型组相比,电针组大鼠兴奋症状明显减少;IC和CP内c-Fos阳性细胞数明显减少。结论电针对氯胺酮滥用所致的IC和CP内c-Fos表达增强具有下调作用。 Objective To determine whether electroacupuncture influences the expression of c-Fos in insular cortex( IC) and caudate putamen( CP) of the rats with ketamine abuse. Methods A total of 32 Sprague-Dawley rats were randomly divided into normal group, normal saline( NS) group, model group and EA group. The ketamine abuse model was established by intraperitoneal administration of ketamine( 100 mg / kg), once a day for 7 days. After the modeling of rats, EA( 2Hz, 1ms, 1m A) was applied to unilateral' Sanyinjiao'( SP6) and 'Zusanli'( ST36) for 30 min, once daily for 7days. The level of c-Fos in IC and CP was detected by immunohistochemistry. Results Compared with the normal group or NS group, rats of the model group showed excitement symptoms such as tail upturning, body twisting, rearing, leaping, circular motion and legs in the air; and the level of c-Fos in IC and CP was significantly increased. Compared with those of the model group, excitement symptoms were significantly reduced and the expression of c-Fos in IC and CP was significantly decreased in the EA group. Conclusion Ketamine abuse can promote c-Fos expression in IC and CP, and EA can down-regulate such expression and reduce the excitement symptoms of ketamine abuse in rats.
