miR-31促进脊髓损伤后Nestin和NSE表达 被引量:5

摘要 目的对脊髓损伤的miR-31转基因小鼠继发性损伤的恢复状况进行了解,进一步了解和探究miR-31在脊髓损伤修复中起到的促进作用。方法使用Impactor M-Ⅲ脊髓撞击器分别对FVB小鼠和miR-31转基因小鼠进行击打,成功建立小鼠脊髓损伤模型。术后于3d、7d、14d、21d进行BBB评分、H E染色、免疫荧光检测和荧光定量PCR检测。结果两组小鼠在制作脊髓损伤模型后,HE染色结果恢复趋势虽大体相同,但BBB评分显示在7d、14d,miR-31转基因小鼠的脊髓损伤恢复状况优于FVB小鼠。免疫荧光和PCR检测显示:Nestin在14d、21d两个时间点上miR-31转基因小鼠高于FVB小鼠;miR-31转基因小鼠NSE的表达量高于FVB小鼠。结论 miR-31在脊髓损伤后可能发挥着促进神经干细胞再生,改善脊髓损伤模型脊髓内微环境,促进脊髓损伤的恢复的作用。 Objective To understand the recovery situation of spinal cord injury in miR-31 transgenic mice, and to further explored the auxo-action effect of miR-31 on spinal cord injury. Methods Impactor M-III was used to induce spinal cord injury in FVB mice and miR-31 transgenic mice. BBB score, HE staining, immunofluorescence and fluorescence quantitative PCR detection were used at 3d, 7d, 14 d and 21 d. Results HE staining results showed that the recovery trend was roughly the same, but there were still differences between the two groups. The degree of recovery in miR-31 transgenic mice was apparently better than that of FVB mice at7 d and 14 d after operation( P < 0. 05). Immunofluorescence and PCR showed more expression of nestin and NSE in miR-31 transgenic mice than in FVB mice at 14 d and 21 d. Conclusions After spinal cord injury, miR-31 may improve the microenvironment to promote recovery.
