摘要
目的了解抑癌基因Keap1对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞增殖能力的影响。方法用携带EGFP-Keap1慢病毒感染NSCLC细胞系A549细胞,以空载EGFP慢病毒感染的A549细胞为对照;采用细胞计数法建立细胞生长曲线和倍增时间,流式细胞仪检测细胞周期;建立BALB/c裸鼠荷瘤鼠NSCLC模型,观察肿瘤大小,对肿瘤组织切片进行Ki67免疫荧光染色。结果过表达Keap1明显抑制细胞生长,显著延长细胞倍增时间,阻滞细胞周期阻滞,并明显抑制荷瘤鼠肿瘤的生长,降低肿瘤组织Ki67阳性细胞。结论抑癌基因Keap1具有明显抑制NSCLC细胞增殖的作用。
Objective To explore the mechanism of tumor suppressor gene Keapl in the cell proliferation of non-small cell lung cancer(NSCLC).Methods Cells with Keapl overexpression were generated using lentiviral infection.The protein overexpression of Keapl in cells was confirmed by Western blot.Cell proliferation was observed using cell growth curve and doubling time.Cell cycle was examined by flow cytometry(FCM).Furthermore,the influence of Keapl on cell proliferation was observed in vivo by xenograft models.The growth of tumor was evaluated by tumor volume and Ki67 staining.Results Proliferation of A549 was evidently reduced in the case of Keapl overexpression,which was reflected by cell growth curve and doubling time.Similar inhibition was observed in tumor models in vivo,with significantly smaller tumor size in Keapl overexpression than that in the control group.Ki67 staining similarly showed lower proliferation rates in tumor with Keapl overexpression than in the control group.Conclusion The in vivo and in vitro results suggest that Keapl suppresss the proliferation of non-small cell lung cancer cell.
