骨肉瘤中CD133和ALDH1免疫组织化学表达和血管生成增加 被引量:3

摘要 目的检测肿瘤干细胞标记物CD133和ALDH1在骨肉瘤(osteosarcoma,OS)中的免疫组织化学表达及其与微血管密度(microvessel density,MVD)的关系,并分析它们与骨肉瘤患者临床病理参数之间的关系。方法通过免疫组织化学Elivision^(TM)plus法检测100例人骨肉瘤标本和20例骨软骨瘤标本(作为对照组)中CD133、ALDH1和CD34蛋白(微血管标志物)的表达。结果在OS组织中CD133和ALDH1的免疫组织化学染色阳性率明显高于骨软骨瘤组织。CD133和ALDH1的阳性表达率与OS患者术后局部复发与否、肺部转移与否、Enneking分期高低均呈正相关;与OS患者的性别、年龄、肿瘤位置、肿瘤大小及组织学类型等无关。MVD计数与OS患者术后局部复发与否、肺部转移与否及Enneking分期均呈正相关。Spearman相关分析发现,CD133和ALDH1的阳性率及MVD计数之间均呈正相关。结论 CD133和ALDH1的表达升高参与OS的浸润、局部复发与转移过程,并可能促进OS组织中的血管生成。 Objective Objective To explore the expression of CD133 and ALDHl and the scores of microvessl density(MVD)in osteosarcoma(OS) and their relationship to clinic pathological parameters.Methods Immunostaining of CD133,ALDH1 and CD34 was performed in 100 cases of osteosarcoma.20 cases of osteochondroma were used as controls.Results The positive rates of CD133 and ALDH1 expression in OS and controls were 47.0%,52.0%vs.5.0%,5.0%,respectively,with significant differences between the two groups.The expression of CD133 and ALDH1 was significantly related with local recurrence,lung metastasis,and Enneking stage of OS patients.There was no significant association between the expression of CD133 or ALDH1 and such characteristics as gender,age,tumor location,tumor diameter,and histological type.The scores of MVD were significantly related with local recurrence,lung metastasis,and Enneking stage of OS patients.Spearman analysis showed that there was a positive association among the expression of CD133,ALDH1 and the scores of MVD.Conclusion High expression of CD133 and ALDHI is involved in the process of invasion,local recurrence,and metastasis of OS,and may promote angiogenesis.
