热化疗诱导胶质瘤细胞凋亡的实验研究 被引量:11

C_6 glioma cell apoptosis induced by hyperthermia combined with antineoplastic agents in vitro

摘要 目的 研究热化疗对C6细胞凋亡的影响 ,评价热化疗抑制细胞增殖的价值。方法 将不同浓度的丝裂霉素直接作用于体外培养的C6胶质瘤细胞 ,接种后 2 0h、48h置于 42 .7℃恒温箱中加温 2次 ,每次 3 0min。观察丝裂霉素对C6细胞的抑制率、生长曲线的影响及热疗、化疗、热化疗对C6细胞凋亡的诱导作用。结果 丝裂霉素在 1.5 μg/ml浓度时杀伤作用达高峰 (P <0 .0 0 1) ,再加大浓度并不增加杀伤作用 (P >0 .0 5 ) ;透射电镜、FCM、TUNEL均观察到热疗、化疗、热化疗诱导细胞凋亡的作用 (P <0 .0 0 1) ;热化疗有协同作用 (P <0 .0 0 1)。结论 ①热疗、化疗、热化疗均抑制C6细胞增殖。②热疗、化疗、热化疗诱导细胞凋亡为其抗癌机制之一。③热疗能提高化疗药物的细胞毒性作用。 Objective To study whether hyperthermia, chemotherapy and thermochemotherapy can inhibit the growth and promote apoptosis of C 6 glioma cell. Methods Experiments were performed as follows:Mitomycin C(MMC)of different concentrations (0.5~2.5 μg/ml) were directly applied to C 6 glioma cell line cultivated in vitro. Then the effects of hyperthermia, chemotherapy, and thermochemotherapy on apoptosis of the C 6 glioma cells induced by the drug were observed and analyzed. Results The killing effect of MMC on C 6 glioma cells peaked at the concentration of 1.5 μg/ml( P <0.001). Transmission electron microscopy, FCM and TUNEL examination observed apoptosis of C 6 glioma cells in every group( P <0.001), and remarkabel synergistic effects between hyperthermia and the drug ( P <0.001). Conclusion Hyperthermia,chemotherapy and thermochemotherapy can all produce a growth inhibiting effect on C 6 glioma cell; Induction of apoptosis of C 6 glioma cells can be achieved by hyperthermia and chemotherapy; Hyperthermia in combination with MMC would provide a greater cytotoxic effect on C 6 glioma cells.
