摘要
目的 探讨高压氧 (HBO)对缺血再灌注小鼠脑组织中细胞因子和血脑屏障的影响。方法 复制清醒小鼠脑缺血再灌注模型 ,并于再灌注期间行 0 .2 5MPa (ATA)HBO治疗 5次 ,在处死动物前 1h经尾静脉注射 2 %伊文思蓝 (Evans) ,采用ABC -ELISA法检测脑组织中IL -1β、TNF -α、IL -6、IL -8的含量及分光光度计法检测脑组织中Evans蓝的含量。结果 脑缺血再灌注组脑组织中细胞因子及Evans蓝的含量较假手术组显著升高 (P <0 .0 5 )。高压氧组与假手术组相比细胞因子及Evans蓝的含量变化不显著 (P >0 .0 5 )。HBO +缺血再灌注组脑组织中细胞因子及Evans蓝的含量较缺血再灌注组显著降低 (分别为P <0 .0 5 ,P <0 .0 1)。结论 HBO可明显减少脑缺血再灌注损伤中脑部细胞因子的含量 ,降低血脑屏障的通透性 ,而对正常脑组织的作用很小。
Objective To investigate the effect of hyperbaric oxygen (HBO) on cytokines in the brain tissues and blood-brain barrier after cerebral ischemia-reperfusion in mice. Methods Using conscious mice cerebral ischemia-reperfusion models, 0.25MPa (ATA) HBO were applied 5 times during the reperfusion period, and an injection of 2% Evans blue into tail veins was conducted an hour before the animals were sacrificed. The levels of IL-1β,TNF-α,IL-6,IL-8 and content of Evans blue in the brain tissue were examined by using ABC-ELISA method and spectrophotometry. Results The levels of the cytokines mentioned above and content of Evans blue were successively increased ( P >0.05) in the ischemia referfusion group as compared with those in the sham surgery group. The levels of cytokines and content of Evans blue in brain in the HBO group are similar to those in the the sham surgery group ( P > 0.05 ). The levels of cytokines and content of Evans Blue in the HBO+ischemia-reperfusion group were successively decreased as compared with those in the ischemia-reperfusim group ( P >0.05, P <0.01). Conclusion HBO ( 0.25 MPa 5 times) has significant effect of reducing the levels of cytokines and the permeability of blood-brain barrier, but has little effect on normal brain tissues.
