摘要
目的对青岛胶东国际机场捕获的2只蝇类进行形态和DNA条形码鉴定。方法对捕获的蝇类,先进行形态学鉴定,然后利用DNA条形码技术,以COI基因为目的基因进行扩增和测序,分析物种同源性并构建系统进化树。结果2只蝇类样本形态学鉴定为花蝇科,无法鉴定到种。通过DNA条形码鉴定和序列比对,2只蝇样本的序列与BOLD数据库中天仙子泉蝇Pegomya hyoscyami的COI基因上一段长达658 bp的序列同源性高达100%,判断为天仙子泉蝇。结论在青岛机场捕获的蝇类为天仙子泉蝇,是青岛口岸首次发现。
Objective To identify the unknown flies collected at Qingdao Jiaodong International Airport with DNA barcoding and morphology methods.Methods Morphological identification was conducted,and the COI gene was amplified by using DNA barcoding technology.The sequence was blast and analyzed.Results It was an Anthomyiidae sp.by morphological identification.The sequence of the DNA barcode of it was 100%identical to the 658 bp published sequence with the COI gene of Pegomya hyoscyami in BOLD.It was showed that the unknown fly species were identified as Pegomya hyoscyami.Conclusion Science and technology novely search result showed that it is a non-recorded species in Qingdao.
