国境口岸病原体检测新方向—重组酶聚合酶扩增技术 被引量:6

New direction for pathogens detection at national frontier ports:recombinase polymerase amplification technology

摘要 作为一项快速、高效、便捷的等温核酸扩增技术,重组酶聚合酶扩增(recombinase polymerase amplification,RPA)技术运用在病原体检测研究中,表现出灵敏度高、特异度好、用时短、应用广泛、操作简单等优势,有望应用于口岸病原体现场快速检测。本文在对比几种等温核酸扩增技术的基础上,重点介绍RPA技术原理和方法,以几种国境口岸重点关注病原体为例,探讨RPA在病原体检测、生物反恐、移动实验室方面的研究进展,为口岸卫生检疫工作提供新的技术参考和方向。 As a rapid,efficient and convenient isothermal nucleic acid amplification technology,recombinase polymerase amplification(RPA)had been used in pathogen detection,with the advantages as high sensitivity and specificity,short reaction time,wide application,simple operation.RPA is applicable to the pathogens detection at national frontier ports.Based on the comparison of several isothermal nucleic acid amplification techniques,this paper focused on the principles and operation of RPA technology,and discussed the application of RPA in pathogen detection,antibioterrorism,and mobile laboratory with several important pathogens as examples.The research progress of RPA pro-vides a new technical reference and direction for the health quarantine at national frontier ports.
