红王子锦带茎段离体培养基的筛选

Screening of Media for the Stem-segment in Vitro Culture of Weigela florida cv.Red Prince

以红王子锦带带腋芽的茎段为外植体进行离体培养基的筛选。结果表明 :红王子锦带的离体培养以MS、WPM为基本培养基 ,附加适当比例的细胞分裂素(0.1～1mg·L-16 -BA)和生长素(0.01～0.1mg·L-1NAA)为宜。本试验采用方差分析及多重比较方法对试验数据进行处理 ,筛选出红王子锦带各培养阶段最佳培养基分别为:(1)外植体诱导培养基(A2) :MS +6 -0.1mg·L -1BA +0.01mg·L -1NAA ;(2)分化及继代培养基(B3) :MS +0.5mg·L -16 -BA +0.05mg·L -1NAA ;(3)生根培养基(C2) :WPM +0.4mg·L -1NAA。

The stem-segments with axillary buds of Weigela floridacv. red prince were used as the explants to sceen the suitable media in vitro culture.The results indicated that MS or WPM as basic medium,added suitable proportion cytokinin(0.1-1mg·L-16-BA)and auxin(0.01-0.1mg·L-1NAA)was appropriate.Through variance analysis and multiple comparison,the appropriate media for different culture stages were determined as:(1)for derivation A2:MS+0.1mg·L-16-BA+0.01mg·L-1NAA;(2)for differentiation and subculture B1:MS+0.5mg·L-16-BA+0.05mg·L-1NAA;(3) for rooting C2 :WPM+0.4mg·L-1NAA.
