摘要
目的 了解羟基喜树碱 (HCPT )对内皮细胞生长的影响 ,进而了解其对血管生成的抑制作用。方法 ECV 3 0 4细胞体外培养并检测不同浓度HCPT对细胞增殖率、形态、粘附功能的影响 ,采用流式细胞仪测定细胞凋亡指数和细胞周期。结果 终浓度为 4.0、7.5、15 .0、3 0 .0、60 .0ng/ml的HCPT对ECV 3 0 4的增殖抑制率分别为 6.7%、3 7.9%、69.5 %、87.4%、91.0 % ;7.5ng/ml和 15 .0ng/ml浓度处理的细胞凋亡率分别为 2 0 .0 %和 3 5 .1% ;细胞的形态和贴壁能力均有明显改变。 结论 HCPT可抑制内皮细胞增殖 ,并能使其形态改变 ,粘附功能下降 ,显示了其抗血管生成的潜能。
Objective To reveal the antiendothelial capability of 10 Hydroxycamptopthecin(HCPT),furthermore to reveal the antiangiogenic capability of HCPT.Methods ECV304 was cultured in medium supplemented with each HCPT dose(4.0 ng/ml,7.5 ng/ml,15.0 ng/ml,30.0 ng/ml,60.0 ng/ml).The cell number was estimated,the rate of inhibition was measured,and morphologic change was observed;the apoptotic rate of cell was tested by a flow cytometry.Results The rate of inhibition (6.7%,37.9%,69.5%,87.4%,91.0%) was correlated with the concentration of HCPT;the apoptotic rate (20.0%,35.1%) was significantly different from the normal(2.2%)( P <0.001),cell morphology and adhesive capability were significantly changed.Conclusion The study has shown the antiendothelial and antiangiogenic capability of HCPT in vitro.
