摘要
采用 ODS柱 ,以 0 .0 2 %磷酸二氢钾溶液 -甲醇 (85∶ 15 ,p H 3.0 )为流动相 ,流速 1m l/ m in,检测波长为2 4 9nm,阿昔洛韦作内标 ,测定小鼠血清中 8-羟基鸟嘌呤的浓度。该法日内及日间 RSD均小于 5 % ,最低检测限2 ng。
A HPLC method for determination of 8-hydroxyguanine in mice serum was established. An ODS column (200×4.6 mm, 5 μm) was used, with the mobile phase of 0.02% potassium dihydrogen phosphate solution-methanol (85∶15, pH 3.0) at flow rate of 1.0 ml/min. The detection wavelength was 249 nm. Aciclovir was used as internal standard. The RSD of intra- and inter- day was less than 5%. The detection limit was 2 ng.
