摘要
目的评价金属螯合剂去铁胺(DFO)、乙二胺四乙酸(EDTA)与美罗培南联合应用,对耐碳青霉烯类铜绿假单胞菌(CRPA)和耐碳青霉烯类肺炎克雷伯菌(CRKP)的体外抗菌效应。方法用纸片扩散法测定细菌对美罗培南的敏感性,用棋盘法测定美罗培南单独应用和联合EDTA和去铁胺后的最低抑菌浓度(MIC)。结果美罗培南与EDTA联合应用后对铜绿假单胞菌、肺炎克雷伯菌的MIC_(50)、MIC_(90)较单用维持不变或下降1~4倍,其MIC分布较美罗培南单用差异有统计学意义(P <0. 05),累积抑菌百分率曲线明显左移;美罗培南与去铁胺联合应用后对铜绿假单胞菌、肺炎克雷伯菌的MIC_(50)、MIC_(90)较单用上升2~4倍,其MIC分布较美罗培南单用差异有统计学意义(P <0. 05),累积抑菌曲线右移。结论与金属螯合剂EDTA联合应用,增强了美罗培南对耐碳青霉烯类铜绿假单胞菌和肺炎克雷伯菌的体外抗菌作用,但与去铁胺联合应用后抗菌活性降低。
Objective To investigate the combined effect of meropenem with metal-chelator Ethylenediaminetetraacetic acid disodium salt(EDTA)and desferrioxamine(DFO)on carbapenem-resistant Pseudomonas aeruginosa(CRPA)and carbapenem-resistant Klebsiella pneumoniae(CRKP).Methods Antimicrobial susceptibility testing was carried out according to a unified protocol using Kirby-Bauer method.The checkerboard assay was used to determine the minimum inhibitory concentration(MIC)of mero-penem by combination of EDTA and DFO.Results EDTA reduced the MIC of meropenem against CRPA and CRKP by 1-4 times.The cumulative inhibition ratio curve of mero-penem and EDTA shifted to the left.However the DFO increased the MIC of meropenem against CRPA and CRKP by 2-4 times.Conclusion The combination of EDTA enhanced the antibacterial activity of meropenem against CRPA and CRKP in vitro but the results in DFO were reversed.
