红景天苷对低氧性肺动脉高压大鼠肺组织炎性因子表达的影响 被引量:13

Effect of salidroside on the expression of inflammatory factors in lung tissue of rats with hypoaerobic pulmonary arterial hypertension

摘要 目的研究红景天苷对低氧性肺动脉高压大鼠肺组织炎性因子表达的影响。方法将40只SD大鼠随机分为对照组、模型组及低、高剂量实验组,各10只。对照组大鼠正常饲养,模型组及低、高剂量实验组置于常压低氧舱中(氧浓度8%~11%,二氧化碳浓度1%~3%,每日8 h,共4周),低、高剂量实验组入舱前30 min分别腹腔注射红景天苷16,32 mg·kg^(-1),对照组及模型组注射等量0. 9%Na Cl。检测各组干预前后平均肺动脉压,平均颈动脉压及右心室肥厚指数(RVHI);以免疫组化法检测肺组织中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)表达。结果对照组,模型组及低、高剂量实验组的平均肺动脉压分别为(11. 58±1. 97),(28. 93±2. 03),(24. 22±2. 14),(19. 75±2. 01)mm Hg;平均颈动脉压分别为(98. 41±15. 24),(145. 97±16. 99),(102. 16±16. 24),(99. 75±15. 97) mm Hg;右心室肥厚指数(RVHI)分别为0. 21±0. 08,0. 39±0. 12,0. 33±0. 09,0. 25±0. 10; IL-6蛋白阳性细胞比例分别为(1. 02±0. 43)%,(68. 41±3. 51)%,(47. 96±3. 46)%,(27. 98±3. 03)%;TNF-α蛋白阳性细胞比例分别为(5. 17±1. 25)%,(79. 58±4. 76)%,(58. 05±4. 21)%,(29. 45±3. 84)%。低、高剂量实验组分别与模型组比较,差异均有统计学意义(均P <0. 05)。结论红景天苷可降低低氧性肺动脉高压大鼠肺动脉压及颈动脉压,抑制心室重构,改善肺组织病理损伤,并可下调肺组织炎性因子IL-6,TNF-α蛋白表达。 Objective To explore the effect of salidroside on the expression of inflammatory factors in lung tissue of rats with hypoaerobic pulmonary arterial hypertension.Methods Forty SD rats were randomly divided into control group,model group and low-and high-dose experimental group,with 10 rats in each group.Rats in control group were fed normally.Rats in model group and low-and high-dose experimental group were placed in normal pressure hypoxic chamber(oxygen concentration 8%-11%,carbon dioxide concentration 1%-3%,8 hours a day for 4 weeks).Salidroside injection 16,32 mg·kg-1 was intraperitoneally injected in low-and high-dose experimental group 30 minutes before entering the chamber,while saline injection in control group and model group was equal.The average pulmonary arterial pressure,average carotid arterial pressure,right ventricle hypertrophy index(RVHI),the lung tissue pathological morphology was observed by hematoxylin and eosin(HE)stain;the expression of interleukin-6(IL-6)protein positive cell proportion and detected by immunohistochemistry method.Results Of the control group,model group and the low-,high-dose experimental groups,the mean pulmonary artery pressure were(11.58±1.97),(28.93±2.03),(24.22±2.14),(19.75±2.01)mm Hg,respectively;the mean carotid artery pressure were(98.41±15.24),(145.97±16.99),(102.16±16.24),(99.75±15.97)mm Hg;the right ventricular hypertrophy index(RVHI)were 0.21±0.08,0.39±0.12,0.33±0.09,0.25±0.10,respectively;IL-6 protein positive cell proportion were(1.02±0.43)%,(68.41±3.51)%,(47.96±3.46)%,(27.98±3.03)%,respectively;the TNF-αprotein positive cell proportion were(5.17±1.25)%,(79.58±4.76)%,(58.05±4.21)%,(29.45±3.84)%.There were significant differences between the model group and the low-,high-dose experimental groups(all P<0.05).Conclusion Salidroside can reduce the pulmonary arterial pressure and carotid arterial pressure of hypoaerobic pulmonary arterial hypertension rats,inhibit ventricular reconstruction,improve the lung tissue pathological injury,and down-regulate the expression of the lung tissue inflammatory factors of IL-6,TNF-αprotein.
