摘要
目的研究黄芩苷对人胃癌MGC-803细胞凋亡及相关凋亡基因表达的影响。方法人胃癌MGC-803细胞随机分为对照组(0.9%NaCl)和低、中、高剂量实验组(80,120,160μmol·L-1黄芩苷)。给药24 h后,用Annexin VFITC/PI双染法检测各组细胞凋亡情况;给药48 h后,用蛋白免疫印迹法检测各组人胃癌MGC-803细胞凋亡相关基因p53及B细胞淋巴瘤/白血病-2(Bcl-2)表达情况。结果对照组,低、中、高剂量实验组总凋亡率分别为(2.03±0.41)%,(18.97±3.04)%,(33.96±4.84)%,(54.83±5.92)%;p53相对表达量分别为0.12±0.01,0.32±0.06,0.85±0.10,1.32±0.25,Bcl-2蛋白相对表达量1.52±0.24,1.02±0.11,0.98±0.06,0.85±0.02。低、中、高剂量实验组分别与对照组比较,差异均有统计学意义(均P <0.01)。结论黄芩苷可通过上调人胃癌MGC-803细胞p53蛋白表达,下调抗凋亡蛋白Bcl-2表达,有效促进细胞凋亡,抑制肿瘤进展。
Objective To explore the effect of baicalin on the human gastric carcinoma MGC-803 cell apoptosis and the expression of apoptosis related genes.Methods The human gastric carcinoma MGC-803 cells were randomly divided into control group(0.9%NaCl)and low-/middle-/high-dose experimental group(80,120,160μmol·L-1 baicalin).At 24 h after treatment,the apoptosis in each group were detected by Annexin V-FITC/PI double staining.At 48 h after treatment,the expression of apoptosis-related genes p53 and B-cell lymphoma/leukemia-2(Bcl-2)of human gastric carcinoma MGC-803 cell was measured by Western blot.Results There were statistically significant differences between the control group and the low-/middle-/high-dose experimental group in the apoptotic rates[(2.03±0.41)%vs(18.97±3.04)%,(33.96±4.84)%,(54.83±5.92)%]and the relative expression of p53 protein(0.12±0.01 vs 0.32±0.06,0.85±0.10,1.32±0.25),the relative expression of Bcl 2 protein(1.52±0.24 vs 1.02±0.11,0.98±0.06,0.85±0.02)(P<0.01).Conclusion Baicalin can promote the apoptosis of human gastric carcinoma MGC-803 cell effectively through the up-regulation of p53 protein and the down-regulation of anti-apoptoasis protein Bcl-2,and inhibit the tumor progression.
