摘要
目的探讨番荔枝内酯单体squamocin对胃癌MGC803细胞增殖,侵袭的影响及机制。方法人胃癌MGC803细胞分为低、高剂量实验组和对照组,每组设置8个复孔,实验组细胞分别以1,10μg·L-1squamocin处理,对照组细胞则以等量0.9%NaCl处理。用噻唑蓝(MTT)法检测MGC803细胞增殖抑制情况,用Transwell小室实验检测MGC803细胞侵袭能力,用蛋白免疫印迹法检测MGC803细胞非肌性肌球蛋白重链9基因(MYH-9)及侵袭蛋白表达情况。结果干预24 h,低、高剂量实验组MGC803细胞增殖抑制率分别为(39.76±1.54)%,(58.49±2.86)%。对照组,低、高剂量实验组相对侵袭指数分别为(100.00±0.00)%,(79.05±0.18)%,(52.47±0.15)%;MYH-9蛋白相对表达量分别为1.06±0.23,0.89±0.14,0.59±0.05,基质金属蛋白酶-2(MMP-2)蛋白相对表达量分别为1.56±0.17,1.12±0.09,0.82±0.04,基质金属蛋白酶-9(MMP-9)蛋白相对表达量分别为0.98±0.08,0.82±0.05,0.64±0.07。低、高剂量实验组分别与对照组比较,差异均有统计学意义(均P <0.05)。结论番荔枝内酯单体squamocin可能通过下调MYH-9及MMP-2,MMP-9蛋白表达,抑制胃癌MGC803细胞增殖、侵袭。
Objective To investigate the effects and mechanism of squamocin on proliferation and invasion of gastric cancer MGC803 cells.Methods Human gastric cancer MGC803 cells were divided into experimental-L/-H groups and control group.Each group was provided with8 multiple pores.The cells in experimental-L/-H groups were treated with 1 and 10μg·L-1 squamocin respectively,while those in control group were treated with the same amount of normal saline.The proliferation inhibition of MGC803 cells was detected by MTT assay,the invasion ability of MGC803 cells was detected by Transwell chamber assay,and the expression of non myosin heavy chain 9(MYH-9)and invasive protein in MGC803 cells was detected by Western blot(WB).Results After 24 hours of intervention,the inhibitory rates of MGC803 cell proliferation in experimental-L/-H groups were(39.76±1.54)%and(58.49±2.86)%,respectively;the relative invasive indexes in control group and the experimental-L/-H groups were(100.00±0.00)%,(79.05±0.18)%,(52.47±0.15)%,respectively;the relative expression of MYH-9 protein were 1.06±0.23,0.89±0.14,0.59±0.05,respectively;the relative expression levels of matrix metalloproteinase-2(MMP-2)protein were 1.56±0.17,1.12±0.09,0.82±0.04,respectively;the relative expression levels of matrix metalloproteinase-9(MMP-9)protein were 0.98±0.08,0.82±0.05 and0.64±0.07,respectively.There were statistically significant differences between the control group and the experimental-L/-H group(P<0.05).Conclusion Annona lactone monomer Squamocin can inhibit the proliferation and invasion of gastric cancer MGC803 cells by downregulating the expression of MYH-9,MMP-2 and MMP-9 protein.
