红芪多糖对糖尿病心肌病db/db小鼠心肌纤维化的抑制作用 被引量:11

Inhibition of astragalus polysaccharide on the myocardial fibrosis of db/db mice with diabetes cardiomyopathy

摘要 目的观察红芪多糖对糖尿病心肌病db/db小鼠心肌纤维化的抑制作用。方法将45只雄性db/db小鼠随机分为模型组、实验组和阳性对照组,每组15只。模型组予以灌胃0.9%氯化钠,阳性对照组予以灌胃罗格列酮4 mg·kg-1,实验组予以灌胃红芪多糖200 mg·kg-1,每日1次,共8周,另选15只同品系非糖尿病db/m小鼠为正常组,予以灌胃等量0.9%氯化钠。用Masson染色检测小鼠心肌胶原纤维化程度,以蛋白质印迹法检测心肌组织凋亡相关蛋白表达。结果正常组、模型组、实验组及阳性对照组小鼠胶原纤维阳性染色相对面积分别为(3.28±1.03)%,(35.97±1.27)%,(20.33±1.09)%,(18.96±1.05)%;心肌组织Bcl-2相关X蛋白(Bax)蛋白相对表达量分别为0.59±0.04,0.87±0.03,0.52±0.04,0.54±0.02;B淋巴细胞瘤-2(Bcl-2)蛋白相对表达量分别为0.93±0.05,0.46±0.03,0.65±0.02,0.68±0.03;胱天蛋白酶-3(Caspase-3)蛋白相对表达量分别为0.61±0.02,0.99±0.03,0.54±0.03,0.52±0.02。实验组和阳性对照组分别与模型组比较,差异均有统计学意义(均P<0.05)。结论红芪多糖可通过调节Bcl-2,Caspase-3和Bax蛋白表达,延缓糖尿病心肌病db/db小鼠心肌纤维化。 Objective The inhibition effect analysis of astragalus polysaccharide on the myocardial fibrosis of diabetes cardiomyopathy db/db mice.Methods Forty-five male db/db mice were randomly assigned to model group,test group,positive control group,with 15 cases in each group.The model group was gavaged with 0.9%NaCl,the positive control group was gavaged with 4 mg·kg-1 rosiglitazone,the test group was gavaged with 200 mg·kg-1 astragalus polysaccharide.All groups received the above treatment once daily for 8 weeks.In addition,a total of 15 homologous non-diabetes db/m mice were assigned to normal group,and were gavaged with 0.9%NaCl.The myocardial collagen fibrosis was detected by Masson staining;the expression of apoptosis related proteins in myocardial tissue was detected by Western blot.Results The relative area of collagen positive staining in normal group,model group,test group and positive control group were(3.28±1.03)%,(35.97±1.27)%,(20.33±1.09)%,(18.96±1.05)%,respectively;the relative expression of myocardial tissue Bcl-2 associated X protein(Bax)were 0.59±0.04,0.87±0.03,0.52±0.04,0.54±0.02,respectively;the relative expression of B cell lymphoma/lewkmia-2(Bcl-2)protein were 0.93±0.05,0.46±0.03,0.65±0.02,0.68±0.03,respectively;the relative expression of cysteine proteinase-3(Caspase-3)were 0.61±0.02,0.99±0.03,0.54±0.03,0.52±0.02,respectively.There were statistically significant differences between the model group and the test/positive control group(all P<0.05).Conclusion Astragalus polysaccharide can inhibit myocardial cell apoptosis and then adjust the blood sugar of diabetes cardiomyopathy db/db mice,delay the myocardial fibrosis through regulation the expression of Bcl-2,Caspase-3 and Bax protein.
