摘要
目的观察莪术对胆管结扎致肝纤维化小鼠的影响。方法将小鼠随机分成3组:对照组、模型组和实验组,每组15只。模型组和实验组小鼠采用胆管结扎的方法构建肝纤维化模型。术后2周,实验组小鼠予以灌胃莪术水煎剂溶液50 mg·kg-1,对照组和模型组小鼠则予以灌胃等量0. 9%氯化钠,每日1次,共8周。用试剂盒检测小鼠血清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)、胆红素水平,用蛋白质印迹法检测小鼠肝组织中Gli1、Ptch-1蛋白表达。结果对照组、模型组和实验组小鼠血清GPT分别为(15. 58±2. 37),(1262. 05±220. 19),(612. 89±251. 68) U·L-1;GOT分别为(124. 78±14. 56),(813. 39±310. 39),(174. 06±113. 25) U·L-1;胆红素分别为(6. 24±1. 46),(265. 43±22. 16),(140. 50±33. 19)μmol·L-1;小鼠肝组织中Gli1蛋白相对表达量分别为0. 85±0. 12,1. 56±0. 21,1. 02±0. 06;Ptch-1蛋白相对表达量分别为0. 79±0. 24,1. 63±0. 21,0. 75±0. 11;对照组和实验组分别与模型组比较,差异均有统计学意义(均P <0. 05)。结论莪术可下调Gli1、Ptch-1蛋白表达,降低胆管结扎诱导的肝纤维化小鼠的炎症反应。
Objective To investigate the therapeutic effect of zedoary turmeric on hepatic fibrosis(HF)mice induced by bile duct ligation and its relationship with Hedgehog signaling pathway.Methods Kunming mice were randomly divided into 3 groups:control group,model group and test group,15 mice in each group.The HF model was established in the model group and the test group using bile duct ligation.At 2 weeks after modeling,the test group was given oral administration of zedoary turmeric decoction solution(50 mg·kg-1)once daily for 8 weeks,while the mice in the control group and model group were given equal amount of 0.9%NaCl.The level of glutamic-pyruvic transaminase(GPT),glutamic-oxaloacetic transarninase(GOT),bilirubin in serum of mice were detected by kit,and the expression of Gli1,Ptch-1 protein in liver tissue of mice was detected by Western blotting.Results The parameters of the control group,the model group and the test group were as follows:the levels of GPT were(15.58±2.37),(1262.05±220.19),(612.89±251.68)U·L-1,respectively;the levels of GOT were(124.78±14.56),(813.39±310.39),(174.06±113.25)U·L-1,respectively;bilirubin were(6.24±1.46),(265.43±22.16),(140.50±33.19)μmol·L-1,respectively;the relative expression of Gli1 protein were 0.85±0.12,1.56±0.21,1.02±0.06,respectively,and the relative expression of Ptch-1 protein were 0.79±0.24,1.63±0.21 and0.75±0.11,respectively.There were significant differences between the control/test group and the model group(all P<0.05).Conclusion Zedoary turmeric can down-regulate the expression of Gli1,Ptch-1 protein and reduce the inflammatory response of HF mice induced by bile duct ligation.
