摘要
目的研究天麻多糖对脑瘫幼鼠模型脑组织神经细胞凋亡基因表达及智力影响。方法用分离结扎颈总动脉及低氧处理构建脑瘫幼鼠模型,将构建成功的21只幼鼠随机分为模型组、对照组和实验组,各7只;8只幼鼠仅分离而不结扎颈总动脉,术后不进行低氧处理为假手术组。实验组腹腔注射天麻多糖300 mg·kg-1,对照组腹腔注射神经节苷脂20 mL·kg-1,假手术组和模型组腹腔注射等量生理盐水,每日1次,连续治疗14 d。用Y迷宫实验评估幼鼠智力,以DNA断裂的原位末端标记法(TUNEL)检测幼鼠脑组织神经细胞凋亡,以逆转录-聚合酶链式反应(RT-PCR)检测幼鼠脑组织凋亡基因表达。结果假手术组、模型组、对照组和实验组Y型迷宫实验正确判断次数分别为(19. 23±1. 68),(8. 12±2. 29),(13. 24±1. 21),(15. 62±2. 12)次,脑组织神经细胞凋亡指数(AI)分别为(0. 96±0. 21)%,(19. 45±1. 26)%,(15. 26±2. 01)%,(12. 03±1. 25)%;对照组和实验组比较,差异均有统计学意义(均P <0. 05)。与模型组比较,对照组和实验组幼鼠脑组织B细胞淋巴瘤-2基因(Bcl-2) mRNA相对表达量和Bcl-2相关X蛋白(Bax) mRNA相对表达量差异均有统计学意义(均P <0. 05),且实验组与对照组相比,差异有统计学意义(P <0. 05)。结论天麻多糖可更有效地改善脑瘫幼鼠智力,其机制与其显著抑制脑瘫幼鼠脑组织细胞凋亡有关。
Objective To investigate the effect of gastrodiae rhizoma polysaccharide on the neuronal apoptosis gene expression in brain tissue of cerebral palsy newborn rats and its intellectual impact.Methods The isolated and ligated common carotid arteries and postoperative hypoxia treatment were used to establish the cerebral palsy newborn rats model,the successful construction of 21 newborn rats were randomly divided into model group,control group and test group,each group with 7 rats,8 rats were in sham-operation group,only separated without ligating the common carotid artery,and had no postoperative hypoxia treatment.Test group was intraperitoneal injection of gastrodiae rhizoma polysaccharide 300 mg·kg-1,control group was intraperitoneal injection of ganglioside20 mL·kg-1,sham-operation group and model group were intraperitoneal injection with the same amount of normal saline,once a day for 14 d continuously.The Y maze test was used to evaluate the intelligence of newborn rats,terminal dexynucleotidyl transferase(TdT)-mediated dUTP nick end labeling(TUNEL)method was used to detect brain tissue apoptosis,the expression of apoptosis-related genes in brain tissue was detected by reverse transcription-polymerase chain reaction(RT-PCR).Results The number of correct judgments of Y-maze test in sham-operation group,model group,control group and test group were 19.23±1.68,8.12±2.29,13.24±1.21,15.62±2.12,brain apoptosis index(AI)were(0.96±0.21)%,(19.45±1.26)%,(15.26±2.01)%,(12.03±1.25)%.The number of correct judgments of the Y maze test and AI in control group and test group had significant difference compared with model group(P<0.05).Compared with model group,the relative expression of B cell lymphoma-2(Bcl-2)mRNA and Bcl-2 related X protein(Bax)mRNA relative expression of brain tissue in control group and test group had significant difference compared with model group(P<0.05),and there was significant difference between control group and test group(P<0.05).Conclusion Gastrodiae rhizoma polysaccharide can improve the intelligence of cerebral palsy newborn rats more effectively,and its mechanism is related to its significant inhibition of brain cells apoptosis in cerebral palsy newborn rats.
