鸡骨草总皂苷对化学性及免疫性肝损伤的保护作用 被引量:8

Protective effects of total saponins of Abrus cantoniensis on chemical and immunological liver injury

摘要 目的建立小鼠化学性和免疫性肝损伤模型,研究鸡骨草总皂苷对肝损伤的保护作用。方法通过腹腔注射1%四氯化碳(CCl4)橄榄油溶液制造小鼠化学性肝损伤模型;通过尾静脉注射刀豆蛋白A(Con-A)制造小鼠免疫性肝损伤模型,对照组和低、高剂量实验组分别给予50 mg·kg-1水飞蓟素和50,100mg·kg-1鸡骨草总皂苷,模型组和空白组分别给予等量的生理盐水。检测各组小鼠血清谷草转氨酶(GOT)和谷丙转氨酶(GPT)水平,检测各组小鼠肝组织中超氧化物歧化酶(SOD)与丙二醛(MDA)水平。通过培养小鼠原代肝细胞和脾细胞,用总皂苷药物处理,用CCK-8法测定肝细胞及脾细胞的存活率。结果高剂量实验组能显著降低化学性和免疫性肝损伤小鼠血清中的GPT水平至(11. 45±5. 21),(36. 45±18. 21) U·L-1,降低GOT水平至(21. 63±2. 36),(57. 65±20. 36) U·L-1,降低肝组织中MDA水平至(27. 03±6. 96),(1. 58±0. 75) nmol·mg-1,增加SOD活性至(151. 48±25. 64),(374. 41±97. 62) U·mg-1。鸡骨草总皂苷作用于小鼠肝细胞48 h,肝细胞的存活率均在100%以上,作用于小鼠脾细胞72 h,存活率均在85%以上。结论鸡骨草总皂苷对化学性与免疫性肝损伤具有较好的保护作用,对肝细胞与脾细胞未显示有毒性。 Objective The protective effects of total saponins extracted from Abrus cantoniensis Hance(ACS)on liver injury were investigated by chemical and immunological liver injury models.Methods The model of chemical liver injury in mice was induced by intraperitoneal injection of 1%CCl4 olive oil solution and the model of immunological liver injury in mice was induced by tail intravenous injection of concanaualin A(Con-A).The control group,Exp-L and Exp-H groups were given 50 mg·kg-1 silymarin and 50,100 mg·kg-1 ACS respectively,model group and blank group were given the same amount of normal saline.After intragastric administration,the serumglutamic-oxaloacetic transaminase(GOT),glutamic-pyruvic transaminase(GPT)activity and superoxide dismutase(SOD),malondialdehyde(MDA)levels in mouse liver tissues were detected.Mouse primary hepatocytes and spleen cells were cultured with ACS treatment and the survival rate was determined by CCK-8 method.Results Exp-H group can significantly reduce the levels of GPT to(11.45±5.21),(36.45±18.21)U·L-1 and GOT to(21.63±2.36),(57.65±20.36)U·L-1 in serum of mice with chemical liver injury or immunological liver injury,reduce the level of MDA to(27.03±6.96),(1.58±0.75)nmol·mg-1 and increase the activity of SOD to(151.48±25.64),(374.41±97.62)U·mg-1 in the liver tissue from mice with chemical liver injury or immunological liver injury.ACS was acted on mouse hepatocytes for 48 h and the cell survival rate was above 100%,acted on spleen cells for 72 h,the cell survival rate was above 85%.Conclusion The results suggest that ACS possess good protective effect on chemical and immunological liver injury,and has no significant toxicity to hepatocytes and splenocytes.
