氢溴酸高乌甲素对神经病理性疼痛大鼠疼痛行为学和海马炎性因子的影响 被引量:4

Effects of lappaconitine hydrobromide on pain behavior and hippocampal inflammatory factors in rats with neuropathic pain

摘要 目的研究氢溴酸高乌甲素对神经病理性疼痛大鼠模型的镇痛效果及对海马炎性因子的影响。方法将SD大鼠随机分为假手术组、模型组和实验组,各25只,用坐骨神经慢性压迫损伤法构建神经病理性疼痛大鼠模型,假手术组仅游离坐骨神经但不结扎。建模后实验组鞘内注射15μg·kg-1氢溴酸高乌甲素,假手术组及模型组则注射等量生理盐水,各组均连续干预5 d。检测大鼠机械刺激缩爪潜伏期(PWT),以逆转录聚合酶链反应(RT-PCR)及酶联免疫吸附法(ELISA)检测大鼠海马组织中炎性因子表达水平。结果术后7 d,假手术组、模型组和实验组PWT分别为(37. 85±3. 26),(17. 45±2. 64),(19. 52±2. 45) g;TNF-αmRNA相对表达量分别为1. 50±0. 22,2. 95±0. 41,2. 25±0. 33,IL-6 mRNA相对表达量分别为1. 25±0. 11,2. 06±0. 22,1. 85±0. 16;TNF-α含量分别为(116. 52±13. 85),(425. 16±20. 63),(189. 56±10. 46) pg·mg-1,IL-6含量分别为(89. 25±12. 36),(369. 45±10. 75),(169. 52±15. 42) pg·mg-1;术后14 d,PWT分别为(38. 15±2. 69),(17. 41±3. 05),(20. 56±2. 36) g,TNF-αmRNA相对表达量分别为1. 46±0. 15,1. 89±0. 23,1. 52±0. 17,IL-6 mRNA相对表达量分别为1. 06±0. 05,1. 68±0. 24,1. 46±0. 16,TNF-α含量分别为(112. 05±14. 52),(369. 25±16. 42),(152. 12±10. 41) pg·mg-1,IL-6含量分别为(86. 59±13. 42),(289. 56±22. 65),(126. 45±15. 28) pg·mg-1,组间差异均有统计学意义(均P <0. 05)。结论氢溴酸高乌甲素可有效提高神经病理性疼痛大鼠痛阈,减轻疼痛反应,其作用机制可能与降低海马组织炎性因子相关。 Objective To investigate the effect of lappaconitine hydrobromide on paroxysmal pain and hippocampal inflammatory factors in rats with neuropathic pain.Methods SD rats were randomly divided into sham-operation group,model group and test group,25 rats in each group,neuropathologic pain rat model were built by chronic compression injury of sciatic nerve,rats in sham-operation group were only freed sciatc nerve but not ligated.Test group was intrathecal injected with 15μg·kg-1 lappaconitine hydrobromide after modeling,sham-operation group and model group were injected with equal amount normal saline,each group was intervened for 5 d continuously.The mechanical paw withdrawal threshold(PWT)were detected;reverse transcription polymerase chain reaction(RT-PCR)and enzyme linked immunosorbent assay(ELISA)were used to detect the expression of inflammatory factors in rat hippocampus.Results 7 d after operation,the PWT of sham operation group,model group and test group were(37.85±3.26),(17.45±2.64),(19.52±2.45)g,the relative expression of TNF-αmRNA were 1.50±0.22,2.95±0.41,2.25±0.33,the relative expression of IL-6 mRNA were1.25±0.11,2.06±0.22,1.85±0.16,TNF-αwere(116.52±13.85),(425.16±20.63),(189.56±10.46)pg·mg-1,IL-6 were(89.25±12.36),(369.45±10.75),(169.52±15.42)pg·mg-1;14 d after operation,the PWT of sham operation group,model group and test group were(38.15±2.69),(17.41±3.05),(20.56±2.36)g,the relative expression of TNF-αmRNA were 1.46±0.15,1.89±0.23,1.52±0.17,the relative expression of IL-6 mRNA were 1.06±0.05,1.68±0.24,1.46±0.16,TNF-αwere(112.05±14.52),(369.25±16.42),(152.12±10.41)pg·mg-1,IL-6 were(86.59±13.42),(289.56±22.65),(126.45±15.28)pg·mg-1,all with significant difference(all P<0.05).Conclusion Lappaconitine hydrobromide can effectively increase the pain threshold and alleviate the pain response in rats with neuropathic pain.
