白藜芦醇对糖尿病心肌病大鼠心功能的保护作用 被引量:6

Protective effect of resveratrol on cardiac function of diabetic cardiomyopathy rats

摘要 目的研究白藜芦醇(RSV)对糖尿病心肌病(DCM)大鼠心功能的保护作用。方法将60只6周龄的SPF级SD雄性大鼠随机分为4组,对照组、模型组、低剂量实验组和高剂量实验组,每组15只。模型组用腹腔一次性注射链脲佐霉素(STZ)(60 mg·kg-1)方法构建DCM大鼠模型。建模成功后,对照组、模型组则予以灌胃等体积的0.9%Na Cl,低、高剂量实验组分别予以灌胃RSV5 mg·kg-1,50 mg·kg-1,每日1次,给药10周。用TUNEL法测定细胞凋亡阳性指数(AI),用Western blot检测Bax、Bcl-2、Wnt3a、p-GSK-3p、β-catenin蛋白的表达。结果对照组、模型组和低、高剂量实验组AI分别为5.81±0.81,18.92±1.51,11.32±1.21和8.81±1.32;Bcl-2/Bax分别为分别为1.91±0.13,0.41±0.11,0.62±0.22和0.92±0.21;Wnt3a分别为1.12±0.11,4.42±0.41,2.51±0.41和1.72±0.21;p-GSK-3p分别为1.11±0.21,3.81±0.51,1.91±0.42和1.52±0.32;β-catenin分别为1.12±0.21,4.61±0.51,2.41±0.41和1.62±0.21。对照组的上述指标和模型组比较,差异均有统计学意义(均P<0.05);低、高剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论 RSV可以通过调节Wnt/β-catenin通路保护DCM大鼠的心功能。 Objective To investigate the mechanism of resveratrol(RSV)protecting heart function in diabetic myocarditis rats by Wnt/β-catenin signaling pathway.Methods Sixty 6-week-old SPF SD male rats were randomly divided into 4 groups,control group,model group,low-dose test group and high-dose test group,with 15 rats in each group.The model group was established by intraperitoneal injection of streptozotocin(STZ)(60 mg·kg-1).After the model was successfully established,the control group and the model group were given oral administration of equal volume of 0.9%Na Cl,while the low and high dose test groups were given RSV 5 mg·kg-1and 50 mg·kg-1once daily for10 weeks after STZ(60 mg·kg-1)injection,respectively.The apoptosis index(AI)was determined by TUNEL method,and the expressions of Bax,Bcl-2,Wnt3a,p-GSK-3p andβ-catenin proteins were detected by Western blot.Results AI levels of control group,model group,low and high dose test groups were 5.81±0.81,18.92±1.51,11.32±1.21 and 8.81±1.32,respectively.Bcl-2/Bax expressions were 1.91±0.13,0.41±0.11,0.62±0.22 and 0.92±0.21,respectively.Wnt3a expressions were 1.12±0.11,4.42±0.41,2.51±0.41 and 1.72±0.21,respectively.P-GSK-3p expressions were 1.11±0.21,3.81±0.51,1.91±0.42 and 1.52±0.32,respectively.Theβ-catenin expressions were 1.12±0.21,4.61±0.51,2.41±0.41 and 1.62±0.21,respectively.The differences between the control group and the model group were statistically significant(all P<0.05),while the differences between the low,high dose test groups and the model group were statistically significant(all P<0.05).Conclusion RSV can protect the cardiac function of DCM rats by regulating Wnt/β-catenin pathway.
