维A酸诱导的骨质疏松大鼠模型的构建及评价 被引量:9

Construction and evaluation of retinoic acid-induced osteoporosis rat model

摘要 目的构建与评价维A酸诱导的雌性大鼠骨质疏松动物模型。方法 12周龄SPF级健康雌性SD大鼠20只,随机分为正常组、模型组,每组10只。模型组给予80 mg·kg-1维A酸连续灌胃21 d诱导骨质疏松。灌胃结束后,采集血液样本,分离血清,检测血清中Ca、P、碱性磷酸酶(ALP)含量,采集股骨组织样本观察骨细胞数和骨形态计量学指标的变化,并用病理图像分析系统进行组织形态计量学测定,用于评价维A酸诱导的骨质疏松大鼠模型。结果模型组和正常组股骨的骨小梁厚度分布为(47.6±7.3),(84.8±13.2)μm,骨小梁间距分别为(168.1±15.3),(126.2±15.8)μm,骨小梁面积占比分别为(42.35±10.67)%和(69.10±9.03)%,血清Ca分别(2.5±0.2)和(2.3±0.2)mmol·L-1,血清ALP分别为(85.4±4.3)和(77.6±7.7)U·L-1,差异均有统计学意义(均P<0.05)。结论根据骨形态计量参数及血清骨代谢指标观察结果表明骨质疏松模型构建成功。 Objective To eatablish and evaluate the animal model of osteoporosis induced by retinoic acid in female rats.Methods Twenty healthy female SD rats of 12 weeks age were randomly divided into normal group and model group,10 in each group.The model group was given 80 mg·kg-1of retinoic acid for 21 d to induce osteoporosis.At the end of 21 d,blood samples were collected,serum was separated,serum Ca,P,and alkaline phosphatase(ALP)levels were measured.The femoral bone samples were collected to observe changes in bone cell number and bone histomorphometry,and analyzed by pathological images,and then the retinoic acid-induced osteoporosis rat model was evaluated.Results The trabecular thickness of the femoral in the model and normal groups were(47.6±7.3),(84.8±13.2)μm,and the trabecular spacing were(168.1±15.3),(126.2±15.8)μm,the area of trabecular bone area was(42.35±10.67)%,and(69.10±9.03)%.Serum Ca were(2.5±0.2)and(2.3±0.2)mmol·L-1,serum ALP were(85.4±4.3)and(77.6±7.7)U·L-1,respectively.There were significant differences of those indexes between the model and normal groups(all P<0.05).Conclusion According to the observation of bone morphometric parameters and serum bone metabolism indicators,the osteoporosis model was successfully constructed.
