水蛭对IgA肾病血瘀证大鼠治疗作用的实验观察 被引量:7

摘要 目的目的观察水蛭对IgA肾病血瘀证大鼠肾功能、24 h尿蛋白定量、血液黏度、白介素-6的影响。方法选用6周龄的Wistar雄性大鼠80只,建立血瘀证IgA肾病模型(空白对照组20只除外),造模成功后,随机分为模型组、强的松治疗组、水蛭组,每组20只,给药喂养至14周末,收集24 h尿,采血,行24 h尿蛋白定量、BUN、Scr、IL-6及全血黏度、血浆黏度检测。结果经治疗后,强的松治疗组及水蛭组BUN、Up、IL-6均较模型组显著下降(P <0. 01),强的松治疗组Scr较模型组显著下降(P <0. 05),但水蛭组疗效较强的松治疗组更显著(P <0. 01);强的松治疗组及水蛭组均可显著降低全血黏度及血浆黏度(P <0. 01),水蛭治疗组疗效显著优于强的松治疗组(P <0. 01),水蛭治疗组高切全血黏度及血浆黏度与空白对照组比较,差异无统计学意义(P>0. 05)。结论水蛭可改善IgA肾病血瘀证大鼠全血黏度及血浆黏度,下调IL-6,减少尿蛋白,保护肾功能。继续深入研究以找到水蛭对IgA肾病血瘀证干预机制及作用靶点,可为临床使用水蛭治疗IgA肾病血瘀证提供更丰富的理论依据。 Objective To observe the effect of leech on renal function,24-hour urine protein quantification,blood viscosity and interleukin-6 on treatment of IgA nephropathy rats with blood-stasis syndrome.Method Eighty male Wistar rats with 6 weeks-old were choose to establish the model of IgA nephropathy with blood-stasis syndrome except twenty male Wistar rats in blank control group.The successfully established experimental models were randomly divided as the model group,prednisone treatment group and leech group with twenty rats in each group.The drug was given until the end of 14 weeks.The experiment indexes of 24 hours Urine protein,BUN,Scr,interleukin-6,whole blood viscosity and plasma viscosity were measured after collecting urine of 24 hours and blood.Results The levels of BUN,Up and interleukin-6 in the prednisone group and leech group were significantly lower than those in the model group(P<0.01).The level of Scr in the prednisone treatment group was significantly lower than that of the model group(P<0.05).However,the efficacy of the leech group was more significant than the prednisone group(P<0.01).Both the prednisone treatment group and the leech group could significantly reduce the whole blood viscosity and plasma viscosity(P<0.01),and the efficacy of the leech group was significantly better than the prednisone group(P<0.01).There was no significant difference in the leech treatment group and blank control group in whole blood viscosity and plasma viscosity(P>0.05).Conclusion Leech can protect renal function of IgA nephropathy rats with blood-stasis syndrome via improving whole blood viscosity and plasma viscosity,down-regulating interleukin-6 and reducing urinary protein.Further research is needed to find out the intervention mechanism and target of leech on the treatment of IgA nephropathy rats with blood-stasis syndrome,so as to provide more theoretical basis in clinical application for leech on the treatment of IgA nephropathy with blood-stasis syndrome.
