新疆棉花枯萎病菌的RAPD分析 被引量:11

摘要 将供试新疆棉花枯萎病菌32个代表菌株及3个标准菌株进行RAPD分析,筛选出14个随机引物,共产生252个RAPD分子标记,其中79.4%具有多态性。通过聚类分析将供试菌株划分为5个RAPD组,此结果与常规鉴别寄主反应法划分生理小种的结果基本一致,RAPD分析为传统方法的结果提供了分子水平的证据,从分子水平上证明新疆棉花枯萎菌群体内7号小种占绝对优势。RAPD可作为一种标记手段用于棉花枯萎病菌生理小种及遗传分化的检测。 RAPD markers was used to analysis genetic diversity among 32 isolates of Frusarium oxys-porum f. sp. vasinfectum on cotton in Xinjiang and three standard races. 14 random primers were selected to used RAPD analysis, 252 RAPD markers were obatined, 79. 4% were polymorphisms. Experiment strains were classified 5 RAPD groups by cluster analysis, The result was radically in accordance with the conclusion from the regular response of identification host, RAPD analysis provided the proof of molecular level for the result of traditional method based on pathogenicity differentiation, It proved that the race 7 is predominant in the isolates of Frusarium oxysparum f. sp. vasinfectum on cotton in Xinjiang from molecular level. The result also shows that RAPD technology is a genetic marker method to delect races and genetic diversity of Frusarium oxysporum f. sp. vasinfectum on cotton in Xinjiang.
