正交试验优选苦柏洗剂提取工艺研究 被引量:3

Study on the optimization of the extraction technology for Kubai Lotion by orthogonal experiment

摘要 目的测定苦柏洗剂中盐酸小檗碱的含量,优化苦柏洗剂的提取工艺。方法采用高效液相色谱(HPLC)法对苦柏洗剂中盐酸小檗碱的含量进行测定,色谱柱为C18(150 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液(0 min,20%乙腈;10 min,40%乙腈)为流动相,流速1.0 m L/min,进样量10μL,检测波长265nm,柱温30℃;采用正交试验对苦柏洗剂的提取工艺进行优化。结果盐酸小檗碱的线性回归方程为Y=168 427X-22 546,r=0.999 9,盐酸小檗碱在16.9~84.5μg/m L范围内呈良好线性关系。加样回收率平均值为98.44%,RSD值为1.43%。正交试验结果为加20倍量水,提取2次,每次1.5 h。结论本试验结果为苦柏洗剂的开发提供了实验理论依据,该工艺稳定、可靠,可应用于工业化大量生产。 Objective To optimize the extraction technology for Kubai Lotion by determining the content of berberine hydrochloride. Methods The content of berberine hydrochloride in Kubai Lotion was determined by HPLC. The sample was separated on the column of C18( 150 mm×4.6 mm,5 μm) which moving phase was acetonitrile and 0.1% phosphoric acid solution( 0 min,20% acetonitrile; 10 min,40% acetonitrile). The flow rate was 1.0 m L / min; the sample size was 10 μL; the UV detection wavelength was 265 nm; the column temperature was 30℃. The extraction technology for Kubai Lotion was optimized by orthogonal experiment. Results The liner equation for berberine hydrochloride was Y = 168 427X-22 546,r = 0.999 9; the calibration curves for berberine hydrochloride was in good linearity in the range of 16.9 ~ 84.5 μg / m L; the average recovery was 98.44% with RSD of 1.43%. The orthogonal experiment result was: adding 20 times water,extracting for 2 times and each time for 1.5 h.Conclusion The study provides experimental basis for the exploitation of Kubai Lotion. The method is stable and reliable,can be used for large-scale industrialized production.
