回生胶囊联合环孢菌素对骨髓增生异常综合征模型大鼠血常规、T淋巴细胞亚群和相关细胞因子的影响 被引量:4

Effects of Huisheng Jiaonang(回生胶囊) combined with cyclosporin on blood routine,T lymphocyte subsets and correlative cytokines in rats model with myelodysplastic syndrome

摘要 目的观察回生胶囊联合环孢菌素对二甲基苯蒽(DMBA)诱导的骨髓增生异常综合征(MDS)模型大鼠血常规、T淋巴细胞亚群和相关细胞因子的影响。方法将60只SD大鼠采用随机数字表法分为空白组、模型组、回生胶囊组、环孢菌素组、联合组5组,除空白组外,其他各组均采用化学诱变剂DMBA静脉注射建立MDS大鼠模型,于实验第14天开始各组灌胃相应药物,连续灌胃30 d。观察各组大鼠的一般情况;检测外周血中白细胞(WBC)、红细胞(RBC)、血小板(PLT)计数和血红蛋白(Hb)含量;检测外周血中T淋巴细胞亚群CD_3^+、CD_4^+、CD_8^+水平;采用ELISA法检测血清γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)含量;采用苏木素-伊红(HE)染色,观察各组大鼠股骨骨髓病理学改变。结果模型组大鼠骨髓病态造血明显,大鼠外周血中WBC、RBC、PLT计数,Hb含量,以及CD_3^+、CD_4^+、CD_8^+水平均明显降低,血清IFN-γ、TNF-α含量均明显升高,与空白组比较差异有统计学意义(P<0.01);各给药组大鼠骨髓病态造血明显改善,外周血中WBC、RBC、PLT计数,Hb含量,以及CD_3^+、CD_4^+水平均明显升高,血清IFN-γ、TNF-α含量均明显降低,联合组大鼠外周血中CD_8^+水平明显升高,差异有统计学意义(P<0.05或P<0.01),且联合组对外周血中WBC、RBC计数,CD_3^+、CD_4^+水平,以及血清IFN-γ、TNF-α含量的改善作用优于回生胶囊组和环孢菌素组(P<0.05)。结论回生胶囊联合环孢菌素可通过改善病态造血,升高外周血象,调节外周血中T淋巴细胞亚群水平,降低血清IFN-γ、TNF-α含量而发挥治疗MDS的作用。 Objective To observe the effects of Huisheng Jiaonang(回生胶囊) combined with cyclosporin on blood routine,T lymphocyte subsets and correlative cytokines in rats model with myelodysplastic syndrome(MDS)induced by dimethylbenzanthracene(DMBA).Methods Sixty SD rats were divided into 5 groups according to random number table as follows:blank group,model group,Huisheng Jiaonang group,cyclosporin group and combination group.Except the blank group,in other groups,rats model were established by intravenous injection of DMBA,a chemical mutagen.The rats in each group were given corresponding medicines by continuous gavage for 30 days from the 14 th day of experiment.The general condition of the rats in each group was observed.The counts of white blood cell(WBC),red blood cell(RBC),platelet(PLT) and hemoglobin(Hb) content in peripheral blood were determined.The levels of CD3+,CD4+ and CD8+ in T lymphocyte subsets in peripheral blood were detected.The levels of serum interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α) was detected by ELISA methods.The pathological changes of femoral bone marrow of rats in each group were observed by hematoxylin-eosin(HE) staining.Results In the model group,the marrow dyshematopoiesis of rats was significant,the counts of WBC,RBC,PLT,Hb content and the levels of CD3+,CD4+ and CD8+ all decreased significantly in peripheral blood of rats,the levels of serum IFN-γ and TNF-α increased significantly,there was statistically significant difference compared with those of the blank group(P<0.01).In each administration group,the improvement of marrow dyshematopoiesis of rats was significant,the counts of WBC,RBC,PLT,Hb content,and the levels of CD3+ and CD4+ all increased significantly,the levels of serum IFN-γ and TNF-α significantly decreased,the level of CD8+increased significantly in peripheral blood of rats in combination group,there was statistically significant difference(P<0.05 or P<0.01),moreover,the improvement of combination group on the counts of WBC,RBC,on the levels of CD3+ and CD4+ in peripheral blood,and on the levels of serum IFN-γ and TNF-α was superior to that of Huisheng Jiaonang group and cyclosporin group(P<0.05).Conclusion Huisheng Jiaonang combined with cyclosporin can play a role in the treatment of MDS by improving the marrow dyshematopoiesis,increasing peripheral hemogram,regulating the level of T lymphocyte subsets in peripheral blood and decreasing the levels of serum IFN-γ and TNF-α.
