缺氧、氧化损伤对培养人TEC整合素α3表达的影响

摘要 目的 观察缺氧、氧化损伤后不同染色程序对培养肾小管上皮细胞 (TEC)整合素α3染色结果的影响。方法 采用免疫荧光激光共聚焦观察乙醇固定及培养活细胞两种实验条件下TEC整合素α3分布的染色结果。结果 正常组TEC整合素α3主要于细胞基底部极性分布 ,缺氧、氧化损伤后出现向细胞侧缘及顶侧分布的趋势 ,活细胞染色仅缺氧组于细胞周缘有阳性荧光染色。结论 缺氧。 Objective To study the effects of anoxia and oxidative stress on the expression of integrin α3 in cultured human tubular epithelial cells (TECs). Methods After anoxia/oxidative stress, immunofluorescence staining and confocal microscopy were used to study the distribution of integrin α3 on the membrane of TECs under 2 experimental conditions, i.e., fixed or cultured living cells. Results The basal polar location of integrin α3 on normal TECs was weakened after anoxia/oxidative stress and tended to diffuse to lateral and apical surface of TECs in fixed condition, but in living cells the immunofluorescence only appeared at the lateral surface of TECs in anoxic groups. Conclusion Exposure of TECs to anoxia/oxidative stress results in the redistribution of integrin α3 from predominantly basal location to the apical and lateral cell surface, and different protocols affect the expression of integrin α3.
