大鼠脑组织切片的显微反射红外光谱

摘要 用电镀法在ITO导电玻璃上制得Co—Ni—Cu金属合金膜,将其用作红外显微反射吸收样品的载玻片,研究了大鼠脑组织切片的化学成分。结果显示:该合金镀层具有较高的反射率,当大鼠脑组织切片在4μm时可得到脑组织中脂类,蛋白质类和磷酸酯类物质的的红外光谱信息。其中在1 740和1 718cm-1处的吸收峰为脂类的特征峰;1 648和1 544cm-1的为酰胺Ⅰ带和酰胺Ⅱ的特征峰;1 234和1 070cm-1为磷酸酯的特征峰。谱图还显示大鼠脑干中的磷酸酯含量高于小脑中的含量。与显微衰减全反射模式相比,反射吸收模式所测得的红外光谱谱峰信号更强。而且,该方法不接触样品,对样品无污染,是真正意义上的无损无接触测量。 The chemical composition of mouse brain tissue was studied by means of infrared reflection micro-spectroscopy.The tissue was mounted onto the Co—Ni—Cu Film electroplated-coated ITO slides.The information of the chemical composition,such as esters,amide and phosphate,can be obtained when the tissue thickness was 4μm.The spectra of the mouse brain tissues showed esters bands at 1 740 cmand 1 718cm-1,amideⅠand amideⅡat 1 648 and 1 544cm-1 and phosphate bands at 1 234 and 1 070cm-1.Also the content of phosphate in brainstem was more than that in cerebellum.Compare with ATR mode,the spectral peak intensity in reflection mode is stranger than that in ATR mode.Also the tissues were not touched when tested in reflection mode with the Co—Ni—Cu Film coated slides,so it can keep the sample free of contamination and non destruction.
