金点阵芯片的SERS超灵敏检测

摘要 SERS技术出现在20世纪70年代,衍生于纳米技术,而今因为其高灵敏度、无损检测和原位测量等优点,被广泛应用于食品安全检测、医药诊断、材料的表征、考古、刑侦等各个领域[1,2]。SERS技术的灵敏度极高,能够达到单分子检测水平,在有害物质检测方面的应用尤为重要。基于利用贵金属层和贵金属纳米粒子产生"热点"增益的思想,制造了金点阵芯片作为活性基底,并以此为基础吸附待检测物质和金纳米粒子,构造三明治结构,实现了对孔雀石绿、罗丹明6G和三聚氰胺等物质的超灵敏检测。这种芯片具有随取随用、操作简单、测量快速便捷、灵敏度高的优点。金点阵芯片为SERS技术的推广提供了一种较好的可行方法,对SERS的研究也具有一定的意义。 SERS was discovered andderived from nanotechnologyin 1 970 s.For the advantages of high sensitive,nondestructive and in-situ testdetection,SERS techniqueis widely used in foodsafetytesting,medical diagnosis,characterizingof material,archaeology,criminal investigationand other fields.SERS technique has high sensitivity,which can reachsingle molecule detection level.It is particularly importantin the application of hazardmaterial detection.Based on theidea thatnoble metal layer and noble metal nanoparticles canconstruct'hot junctions',agold-patterned microarray chip was fabricated.The analyte and the gold nanoparticles were sequentiallydeposited onto the chip to form a sandwich structure and then realized the analyte SERS detection.By using the chip,SERS detection on MG,rhodamine6 Gand melamine were taken.The gold-patterned microarray chipmakes SERS measurement more simple and convenient.This chip supplies an efficient way for SERS promotion and is important to SERS study.
