抗抑郁剂地昔帕明与氟西汀对慢性应激大鼠海马基因表达的影响

摘要 目的 :研究抗抑郁剂相关的基因。方法 :采用不同应激方式交替持续应激 2 0d造成大鼠慢性应激损伤模型 ,提取大鼠海马组织总RNA ,行逆转录聚合酶链反应。PCR产物上样于聚丙烯酰胺凝胶 ,银染法分离差异表达的条带 ,用斑点杂交方法对差异表达片段进行验证 ,将表达量有差异的条带测序。结果 :与慢性应激组相比 ,地昔帕明可以上调微管相关蛋白基因的表达 ,氟西汀可以上调一种神经元肌动蛋白结合蛋白树状棘蛋白的基因表达。结论 Objective:To identify antidepressant related genes.Methods:Rats were subjected to chronic stress for consecutive 20 days by different ways.Total RNA from rat hippocampus was isolated ,reverse_transcribed and amplified by RT_PCR.The PCR products were displayed on polyacrylamide gel by silver staining and the differentially expressed genes were retrieved, cloned and further confirmed by dot_blot hybridization.The differentially displayed cDNAs were sequenced.Results:Desipramine upregulated the microtubule_associated protein (MAP_4) mRNA expression and fluoxetine upregulated the drebrin mRNA expression,compared with the chronic stress group.Conclusions:The results indicate that the desipramine and fluoxetin may play their antidepressant role by upregulating some gene expression which are associated with neuroregeneration in the central nervous system.
