5种虫媒病毒基因组部分核苷酸序列的测定及同源性比较

Part nucleotide sequence determination and homology analysis of five arbovirus genomes

摘要 目的 :从分子水平对引进的 5种虫媒病毒 :马雅罗病毒 (MAY)、西门利克病毒 (SFV)、墨累谷脑炎病毒(MVE)、伊利乌斯病毒 (ILH)和布尼安姆韦拉病毒 (BUN)进行鉴定。方法 :提取病毒基因组RNA ,用逆转录_聚合酶链反应 (RT_PCR)进行 5种虫媒病毒目的片段的cDNA扩增 ,将获得的片段分别克隆于pGEM_T_Easy载体 ,进行测序和同源性分析。结果 :测得的病毒基因组核苷酸序列与相应病毒的序列同源性最高 ,分别为MAY 97% ,SFV 99% ,MVE 99% ,ILH 99%和BUN 98%。结论 :通过对 5种病毒基因组部分核苷酸序列的测定及同源性比较 ,最终将这 Objective:To clone, sequence and analyse the homology of sequences of five arboviruses (MAY, SFV, MVE, ILH and BUN) which belong to Togaviridae, Flaviviridae or Bunyaviridae,respectively.Methods:RNAs of five arboviruses were extracted from infected sucking mice brains. With the RNAs as templates, cDNA fragments containing the regions encoding structure or non_structure protein were obtained by reverse transcription polymerase chain reaction (RT_PCR). The PCR products were cloned into pGEM_T_Easy vector and sequenced. The nucleotide sequences of the five arboviruses were compared with the sequences in GenBank and their homology was analysed.Results:Sequence homology analyses revealed that five arboviruses had the highest homology with their corresponding sequences in GenBank. The corresponding results of homology analyses were MAY 97%,SFV 99%,MVE 99%,ILH 99% and BUN 98%.Conclusion:The five arboviruses were identified by the nucleotide sequence and comparative analysis.
