快速徒手切片法观察谷子和水稻叶片显微结构 被引量:23

Free-hand Section Techniques for Clear Observation of Cell Structures of Millet(Setaria italica) and Rice(Oyrza sativa ) Leaves

摘要 水稻是世界上一半以上人口的主食,且是禾本科植物分子生物学研究的模式植物。谷子是狗尾草属植物,是我国北方重要的杂粮,是典型的C4植物,谷子与水稻基因组共线性很高。对谷子和水稻两种作物的对比研究能够对研发C4水稻提供理论依据。突变体库是研究功能基因组学的重要材料,目前对突变体库的构建技术已经很成熟,但是对目的突变体,尤其是对解剖学方面的突变体材料筛选主要方法是进行石蜡切片,该过程费时费力。本研究旨在改进徒手切片方法对材料进行预筛选,能大大缩减工作量,提高工作效率,该方法得到结构完整、清晰的水稻和谷子叶片徒手切片。 Rice(Oyrza sativa L.) is a staple food feeding more than half of the population in the world. It has become a model plant for the molecular biology study of the Poaceae grass family. Foxtail millet(Setaria italica), a member of the Poaceae grass family, is one of the important grain crops in North China. It is a typical C4 plant and its genome has high co-linearity with that of rice, so the comparison study of millet and rice can provide theoretical basis for research and development of C4 rice. Mutant library is an important material for the study of functional genomics. The techniques and methods for constructing mutant library is mature. However, the screening method to find target mutants, particularly for anatomy related mutants, is usually based on paraffin sectioning, which is time-consuming. This study aimed at optimizing free-hand sectioning for the preliminary screening of anatomy related materials. This method can significantly reduce the labour costs and improve efficiency. Finally, the slices with clear and intact cell structures of rice and millet leaves were obtained by this method.
