摘要
本研究通过对各个品种的DNA池构建及引物设计,扩增其启动子区的部分序列,将PCR产物经纯化后测序。再利用DNAStar和Promoter SCAN等生物在线分析软件对测序结果进行预测分析。共筛选出7个起到关键影响作用的SNPs位点,分别为G-957A、A-880G、A-879G、T-839G、T-821C、C-788T和A-751G。对其进行生物学信息分析得出,突变前后,启动子的位置和启动子内部的转录因子结合位点及Cp G岛均产生了较为重要的变化。结果表明,FASN基因可能对脂肪沉积性状产生重要的影响作用。
The partial sequence of the promoter region was amplified according to construct the varieties of DNA pools and design the primer. After purified, the PCR product was used for detecting sequence. Then, forecasting the result of the sequencing by using the online analysis software such as DNAstar and Promoter SCAN. The seven SNPs by screening has more important influenced. It was G-957 A, A-880 G, A-879 G, T-839 G, T-821 C, C-788 T,A-751 G. The analysis of bioinformatics shows that the change of the mutation was significant, included promoter position and the promoter transcription factor binding sites and Cp G island. The results showed that FASN genes may have important impact on fat deposition properties.
