摘要
本研究以FoxL2基因为研究对象,选取Gen Bank公布的26个物种的FoxL2基因氨基酸序列,代表脊椎动物中5纲15目21科24属,重点分析哺乳动物中该基因的结构与功能相关性以及其系统发育情况。结果表明:26条FoxL2基因序列存在非常保守的forkhead结构域,哺乳类比非哺乳动物多出丙氨酸、甘氨酸及脯氨酸重复区,其中多聚丙氨酸束的缩短可能与Ⅰ型BPES相关,而多聚丙氨酸束的延伸使核内蛋白错位和细胞质聚集导致Ⅱ型BPES。FoxL2含有较多磷酸化位点,推测其在体内的活性可能是通过磷酸化调控的。FoxL2基因氨基酸序列构建的系统发育树中亲缘关系较近的物种能够较好的聚在一起。
In this research we used FoxL2 gene as the research objective,and selected twenty six amino acid sequences of FoxL2 gene from 26 vertebrate species belonging to 24 genera,21 families,15 orders and 5 classes to investigate their structures,functions and phylogenetic among species. The results had demonstrated that conservative forkhead domain structure exists in all 26 FoxL2 gene sequences. The repeated regions for alanine,glycine and proline were found only in mammals but not in non-mammals. Polymers of alanine beam shortening might be associated with BPES type Ⅰand the extension of alanine beam made nuclear protein dislocation and cytoplasm clustering led to BPES type Ⅱ. FoxL2 contained many phosphorylation sites,and its activity was likely to be regulated by phosphorylation in vivo. The phylogenetic tree of FoxL2 gene amino acids sequences showed that species of consanguinity get much closer.
