摘要 在Gen Bank中找到粉质胚乳突变基因Mucronate,构建以bar基因为筛选标记的植物表达载体p CAMBIA3300-Ltp2-Zm MC。通过农杆菌介导的萌动胚转化法将Ltp2-Zm MC基因转入玉米自交系郑58中,用含5 mg/L草胺磷的筛选培养基进行筛选,共获得280株再生植株。经过PCR和RT-PCR鉴定,得到29株RT-PCR阳性植株。结果表明,Zm MC基因在玉米中得到表达。 The mutant gene Mucronate(Mc) of farinaceous endosperm was found according to Gen Bank, and the plant expression vector p CAMBIA3300-Ltp2-Zm MC with the bar gene as the selection marker was constructed.Ltp2-Zm MC gene was transformed into germinating embryo of maize inbred line Zheng 58 by Agrobacterium-mediated transformation, screening with selection medium containing 5 mg/L of PPT to obtain a total of 280 PPT-resistant seedlings. After PCR and RT-PCR identification, 29 plants were positive. The results showed that the Ltp2-Zm MC gene was expressed in maize.
