摘要
鬼针草(Bidens bipinnata L.)是国内一种重要的药用植物,同时也是镉超富集植物。本研究采用OTTOⅠ细胞核提取液配方和PI荧光染料,以鬼针草嫩叶为实验材料,以基因组DNA含量已知的番茄(Solanum lycopersicum)为对照,运用流式细胞术对鬼针草基因组大小进行了测定。结果表明,鬼针草基因组大小约为4 650.674 73 Mbp,按1 pg DNA=0.978×10~9 bp来计算,鬼针草的基因组DNA总量约为9.51 pg。本研究探索了用流式细胞仪分析鬼针草基因组大小的方法,所测得数据可为鬼针草的基因组学研究提供参考。
Bidens bipinnata L. is an important medicinal plant and a Cd-hyperaccumulator in China. In this research, we used the tender leaves of Bidens bipinnata L. as materials, chose Solanum lycopersicum, whose content of genomic DNA was known, as a control, and estimated the genome size of Bidens bipinnata L. by flow cytometry(FCM) using OTTOⅠnucleus extract formula and PI fluorochrome. The results showed that the genome size of Bidens bipinnata L. was about 4 650.674 73 Mbp. The genomic DNA content of Bidens bipinnata L. was about 9.51 pg according to the formula 1 pg DNA=0.978×10~9bp. This research investigated a method of analyzing the genome size of Bidens bipinnata L. by flow cytometry and the measured data would provide a reference for the genomic research of Bidens bipinnata L..
