摘要
探讨竹节参对AD大鼠海马区Drd-2及TNF-α、GFAP表达的影响。60只SPF级SD雄性大鼠,适应性喂养一周后,随机分为6组。除假手术组海马注射生理盐水外,其余5组均注射Aβ1-42以复制AD模型。然后对假手术组及模型组大鼠予以生理盐水灌胃,西药组大鼠予以脑复康悬浮液灌胃,竹节参高、中、低剂量组大鼠分别予以相应浓度竹节参水煎液灌胃,持续4周。完成水迷宫实验后,处死大鼠,取大鼠海马,切片、Western Blotting法检测各组大鼠海马Drd-2、GFAP和TNF-α表达量的变化。Western blotting法观察各组大鼠海马GFAP、TNF-α的表达水平,模型组较假手术组明显升高,各治疗组明显低于模型组,且竹节参高剂量组低于西药组。Drd-2的表达水平,模型组较假手术组明显降低,各治疗组明显高于模型组,且竹节参高剂量组要高于西药组。竹节参能通过提高AD大鼠海马形胶质细胞Drd-2活性,抑制炎症反应。
In order to investigate the effects of Panax japonicus on Drd-2 and GFAP,TNF-αin hippocampus of AD rats,60 male SD rats were randomly divided into six groups after adaptive feeding of 1 week.In addition to injecting normal saline in sham operation group,the other five groups were injected with Aβ1-42 to copy AD model.The rats in the sham operation group and the model group were treated with normal saline gavage.The rats in the western medicine group were treated with piracetam suspension gavage and the rats in low,middle and high dosage Panax japonicus groups were treated with Panax japonicus decoction in different concentrations by gavage,which continued 4 weeks.After the water maze experiment,the rats were killed to obtain their hippocampus,which were later sliced and detected the expression changes of Drd-2,TNF-αand GFAP in each group of rats by Western Blot method.The expression levels of GFAP and TNF-αin the hippocampus of rats were observed by Western Blot method,which in the model group were significantly higher than those in the sham operation group,while these levels in the treatment group were lower than those in the model group,as well as the levels in high dose group was lower than that in western medicine group.The levels of Drd-2 in the model group were significantly lower than those in sham o peration group,and these in treatment group were higher than that in model group and the levels in high dose group was higher than that in western medicine group.Panax japonicus could increase the activity of Drd-2 in hippocampus astrocytes,and reduce the inflammatory response.
