红花多糖可显著抑制HT29结直肠癌细胞增殖 被引量:8

Safflower Polysaccharide could Significantly Inhibit the Proliferation of HT29 Colorectal Cancer Cells

摘要 本研究采用MTT法检测红花多糖对HT29结肠癌细胞增殖的抑制作用,观察大肠癌细胞凋亡的形态,采用Annexin V和PI双染流式细胞仪检测细胞周期和凋亡,采用蛋白质印迹法检测Caspase-3蛋白的表达,试图研究红花多糖对HT29结肠癌细胞增殖的抑制作用及其相关机制。研究结果表明各浓度组的抑制率均明显高于对照组(p<0.05)。随着药物浓度的增加,抑制率更显著,IC50=201.908 mg/L。实验组的浓度分别为160 mg/L、320 mg/L和640 mg/L。HT29细胞周期的影响主要体现在HT29细胞G2/M期和S期。在实验组的浓度为160 mg/L、320 mg/L和640 mg/L时细胞凋亡率显著高于对照组(p<0.05)。在160 mg/L、320 mg/L和640 mg/L组Caspase-3蛋白表达显著高于对照组(p<0.05)。随着浓度的增加,表达量增加。说明红花多糖能显著上调Caspase-3蛋白。本研究初步得出结论:红花多糖能显著抑制HT29结肠癌细胞,其诱导HT29细胞凋亡的机制可能与阻滞细胞G2/M期,S期,及上调Caspase-3蛋白的表达有关。 In order to explore the inhibitory effect of safflower polysaccharide on HT29 colorectal cancer cellproliferation and its relevant mechanism,in this research MTT assay was used to detect the inhibitory effect of safflower polysaccharide on HT29 colorectal cancer cell proliferation and morphology of colorectal cancer cell apoptosis was observed;while cell cycle and apoptosis was assayed using Annexin V and PI double staining flow cytometry,and Caspase-3 protein expression was detected by Western blotting.The results showed that the inhibitory rate in each experimental group with different concentration was significantly higher than that in control group(p<0.05).The inhibitory rate was more significant along with the increasing concentration of the medicine dose-dependently and IC50 was calculated as 201.908 mg/L.The HT29 cell cycle was affected among the concentration to HT29 cells at 160 mg/L,320 mg/L and 640 mg/L in experimental group,respectively,which might block G2/M phase,S phase in HT29 cells;while the apoptosis rate in experimental group with 160 mg/L,320 mg/L and 640 mg/L was significantly higher than that of control group dose dependently(p<0.05).Caspase-3 protein expression in experimental group with 160 mg/L,320 mg/L and 640 mg/L was significantly higher than that of control group(p<0.05).The expression increased along with the increasing concentration,which indicated that safflower polysaccharide could significantly upregulate Caspase-3 protein.Our study preliminarily concluded that safflower polysaccharides can significantly inhibit HT29 colon cancer cells,and its mechanism of inducing HT29 cell apoptosis may be related to blocking G2/M phase,S phase,and up-regulating the expression of caspase-3 protein.
