小剂量阿司匹林所致大鼠胃黏膜损伤ICAM-1表达的研究 被引量:4

摘要 目的:应用小剂量阿司匹林灌服大鼠制备胃黏膜损伤模型,观察小剂量阿司匹林对胃黏膜的损伤、小剂量阿司匹林对胃肠道的危险性,了解细胞间黏附分子(ICAM-1)是否参与小剂量阿司匹林引起的胃黏膜损伤.方法:用5.021mg/kg小剂量阿司匹林灌服大鼠,观察大鼠胃黏膜损伤情况,同时应用免疫组化,检测ICAM-1在胃黏膜细胞的表达.结果:用5.021mg/kg小剂量阿司匹林灌胃后3d实验组开始出现胃黏膜损伤的表现,损伤指数为9.11±0.62,14d损伤达高峰,损伤指数为23.52±0.53.在灌服小剂量阿司匹林的48只实验组大鼠中,有40只出现胃黏膜损伤的改变,损伤的发生率为83.3%.免疫组化结果显示ICAM-1在用药后3d开始表达,7d后逐渐升高,21d达高峰,28d及35d仅有少量表达.实验组用药后与用药前比较ICAM-1表达水平有明显差异;对照组与实验组比较,ICAM-1均明显呈低水平表达;对照组之间ICAM-1表达水平无明显差异.结论:用5.021mg/kg小剂量阿司匹林可以成功地制备胃黏膜损伤动物模型;小剂量阿司匹林可造成胃黏膜损伤、小剂量阿司匹林对大鼠胃黏膜具有一定的危险性;ICAM-1在小剂量阿司匹林引起的胃黏膜损伤的发病机制中具有一定的作用. AIM: To establish a model of gastric mucosal injury inducedwith a low dose of aspirin, and to investigate whether theexpression of ICAM-1 in rat gastric mucosal cells participated inthe pathogenic mechanism of gastric mucosal injury.METHODS:A model of gastric mucosal injury with aspirin(5.021 mg/kg) was established. The changes of gastricmucosal injury was observed. Expression of ICAM-1 proteinof the gastric mucosal cells in SD rats was detected byimmunohistochemistry.RESULTS:Gastric mucosal injury occurred in experimentgroups at the third day after administration of 5.021 mg/kgaspirin and the injury index was 9.11±0.62. Gastric mucosainjury arrived at a peak level at the fourteenth day with theinjury index of 23.52±0.53. Gastric mucosal injury appeared in40 of 48 rats (83.3 %) in experimental groups after admin-istration of aspirin. Immunohistochemical staining showedthat ICAM-1 began to express on the third day afteradministration, peaked at the twenty-first day, decreasedat the twenty-eighth day and the thirty-fifth day. There wasa significant different in the expression of ICAM-1 betweenbefore and after the administration of aspirin in experimentgroups. There was lower expression in the control groupthan that in experiment group. No significant differenceexisted among control groups.CONCLUSION:A model of gastric mucosal injury is successfullyreplicated with a low dose of aspirin, which is harmful togastric mucosa. The expression of ICAM-1 plays an impor- tant role in the mechanism of aspirin-induced gastric mu-cosal injury.
