摘要
目的:建立香苓开胃方中广藿香酮含量的HPLC测定方法。方法:采用高效液相色谱法(HPLC)测定香苓开胃方水煎剂中广藿香酮的含量,色谱柱Extend-C_(18)(4.6mm×250mm,5μm),流动相为乙腈(A)-0.2%的磷酸水溶液(B),梯度洗脱(0~20min,A 55%~60%),进样量:10μL,流速:1mL·min^(-1)。检测波长为310nm,柱温为30℃。结果:广藿香酮在0.00798~0.66500μg(r=1.0000)范围内线性关系良好;平均回收率(N=6)为108.58%,RSD为3.24%。香苓开胃方中广藿香酮含量范围为0.339~1.408 mg·dose^(-1)。结论:本实验方法简便准确,回收率、重复性均能达到要求,可用于香苓开胃方的质量控制。
Object To establish a method for determination of patchoulenone in Xiang Ling appetizing prescription by High Performance Liquid Chromatography(HPLC). Methods: The content of Patchoulenone from the water extract of Xiang Ling appetizing prescription was detected by HPLC. The Extend-C18(4.6 mm×250 mm,5μm)column was used for chromatography. The mobile phase consisted of acetonitrile(A)-0.2%phosphoric(B) with solution gradient elute(0~20 min,A 55%~60%), injection volume was 10 μL, flow velocity was 1.0 mL·min-1, wave length was 310 nm,column temperature was set at 30 ℃. Results: In the range of 0.00798~0.66500μg(r=1.0000), Patchoulenone has a good linear regression, and average recovery was 108.58% with RSD3.24%(N =6). Purity of Patchoulenonein Xiang Ling appetizing prescription is 0.339~1.408 mg·dose-1. Conclusion: This method is simple and accurate, the recovery rate and repeatability can meet the requirements, and can be used for the quality control of Xiang Ling appetizing prescription.
