摘要
表观修饰在基因表达调控、细胞命运决定等众多生物学过程中发挥着关键的调控作用。组蛋白翻译后修饰是一类重要的表观遗传调控机制,调控着染色质层面的遗传信息解读。近年来的研究表明,组蛋白自身突变会导致癌症的发生,突变组蛋白因此被称为'癌组蛋白'(oncohistone)。笔者立足于骨与软组织肿瘤,围绕癌组蛋白的突变及其表观调控机制作一系统综述。现报告如下。
Research on high-frequency mutations of histone coding genes and their carcinogenic properties achieved remarkable results.Histone residues carrying mutations cannot be modified by modifying enzymes,causing changes in genomic stability and gene expression patterns,and ultimately affecting the occurrence of tumors.Therefore,the mutant histone is called'Oncohistone'.High frequency oncohistone mutations have been found in bone and soft tissue tumors such as bone giant cell tumor,chondroblastoma and osteosarcoma.This paper intends to make a systematic review on the involvement of oncohistone mutations in the occurrence and development of bone and soft tissue tumors and the corresponding possible mechanism,and further discuss the progression of accurate tumor treatment based on oncohistone.
