雌激素受体阳性绝经后肥胖乳腺癌的芯片数据分析

摘要 目的:初步探索雌激素受体(ER)阳性绝经后肥胖乳腺癌的相关基因及可能涉及的信号通路。方法:从GEO公共数据库下载含体质指数(BMI)信息的ER阳性绝经后乳腺癌基因芯片数据集(GSE70947),共获得13例肥胖乳腺癌和38例非肥胖乳腺癌的基因表达谱数据,进行基因差异表达谱和信号通路分析,并通过荧光定量PCR法和Western blotting检测法进行相关验证。结果:基因芯片数据分析显示,566个基因在肥胖乳腺癌中出现差异表达,分别有342个基因上调和224个基因下调。GO分析和Pathway分析发现,细胞功能和信号通路可能参与了ER阳性绝经后肥胖乳腺癌的发生发展过程。荧光定量PCR检测结果显示,MMP14、SLC26A3、COL1A2基因mRNA表达情况与芯片分析结果一致。Western blotting检测结果显示,肥胖乳腺癌的COL1A2蛋白相对表达量更高(P<0.05)。结论:本研究分析了ER阳性绝经后肥胖乳腺癌的基因表达谱及可能涉及的信号通路,其中COL1A2基因可能是ER阳性绝经后肥胖乳腺癌的相关基因及潜在的治疗靶点。 Objective:To investigate the gene profiles related to estrogen receptor(ER) positive-breast cancer in postmenopausal obese women and the potential signaling pathways.Methods:A gene microarray dataset of ER positive-breast cancer in postmenopausal obese women(GSE70947) containing data about body mass index(BMI) was downloaded from the GEO public database.The gene expression profiles of breast cancer in 13 obese and 38 non-obese women were obtained,and the differential expression profiles and signal pathways were analyzed.Fluorescence quantitative PCR and Western blotting were used for validation.Results:The microarray data identified 566 differentially expressed genes in breast cancer tissues from obese women,including 342 up-regulated genes and 224 down-regulated genes.GO analysis and Pathway analysis showed that the cell functions and signaling pathways may be involved in the occurrence and development of ER positive-breast cancer among postmenopausal obese women.Fluorescence quantitative PCR showed that the mRNA expression of MMP14,SLC26A3 and COL1A2 was consistent with the findings of microarray analysis.Western blotting showed that the relative expression of COL1A2 protein in breast cancer was higher in obese women.Conclusion:This study analyzes the gene expression profile and potential signaling pathways of ER-positive breast cancer among postmenopausal obese women.The COL1A2 may be a related gene and potential therapeutic target for ER positive-breast cancer in postmenopausal obese women.
