摘要
目的:研究黄连素和环丙沙星对铜绿假单胞菌生物膜的影响。方法:建立体外铜绿假单胞菌野生株P0A1生物膜模型,分别测定黄连素、环丙沙星分别抑制生物膜形成的MIC,以及黄连素和环丙沙星联合用药的抑菌分级指数(FIC)值。结果:当黄连素浓度0.139 g/mL和环丙沙星浓度0.25μg/mL,分别出现明显的抑制生物膜作用。黄连素和环丙沙星联合用药FIC值是0.5。结论:黄连素有抑制生物膜形成的作用,且与环丙沙星联用可有协同抑制生物膜形成作用。在临床工作中治疗耐FQL的PA感染性疾病时,可尝试联合应用黄连素,以起到提高效果、降低成本和减少耐药的效果。
Objective:To study the effects of berberine and ciprofloxacin on P.aeruginosa biofilm.Methods:The biofilm model of P.aeruginosa wild strain P0A1 was established in vitro to determine the MIC of berberine and ciprofloxacin in inhibiting biofilm formation when used alone and the fractional inhibitory concentration(FIC)of berberine and ciprofloxacin when used in combination.Results:When the concentration of berberine was 0.139 g/mL or the concentration of ciprofloxacin was 0.25μg/mL,significant biofilm inhibition was observed.The FIC value of berberine and ciprofloxacin combination was 0.5.Conclusion:Berberine may inhibit biofilm formation.A synergistic inhibitory effect of the biofilm formation is observed with combined use of berberin and ciprofloxacin.In the clinical settings,treatment of fluoroquinolone-resistant Pseudomonas aeruginosa infections may involve combination use of berberine and ciprofloxaxin,so as to improve the therapeutic efficacy,reduce costs and drug resistance.
