摘要
为探讨参附汤(SFT)大鼠血清对人白血病细胞株HL60糖皮质激素受体(GR)及其信使核糖核酸(mRNA)的影响.用动物血清学方法制备SFT血清制剂,取浓度为1×106/ml HL60细胞的细胞悬液培养液150μl加入浓度为20%SFT血清,置于96孔培养板内,培养48h后测定GRmRNA和GR,并以生理盐水血清和含小牛血清细胞培养液作对照.用放射配体结合法分析GR,用Northern blot分析GRmRNA的改变.结果:SFT可增加HL60细胞GR和GRmRNA的含量,生理盐水组则抑制两者的表达.结论:SFT对GR的表达有正向调节作用,结合临床上SFT的抗休克作用,提示SFT的这一作用可能是通过正向调节GR表达后,增加了GC的生物学效应来实现的,其作用点在mRNA转录水平.
In order to explore influence of ' Ginseng and A-conite Decoction' ( GAD) on glucocorticoid receptor (GR) and its messenger NRA of human leukemia clone HL 60 in rat serum, 150(1 culture solution of cellular suspension of HL60 cells in 1(106/ml concentration, after added with, GAD serum in concentration of 20% , was put into 96 - hole culture plate and cultured for 48 hours to determinate GRmRNA and GR, and simultaneously to compare with serum of physiological saline and cellular solution containing calf serum. GAD serum preparation was processed by animal serological methods. GR was analyzed by radioligand integration assay. Changes in GRmRNA were analyzed by Northern blot. The results showed GAD could elevate GR and GRmRNA contents of HL60 cells and two of them were inhibited in the group of physiological saline. The conclusion is that GAD has a positive regulatory function. In combination of the clinical anti - shock function of GAD, the findings indicate that the function of GAD may be realized by the elevation of GC biological effect after positive regulatory GR expression and its function lies in the transcript level of mRNA.
