摘要
目的 探讨反义寡核苷酸 (ASODN)对内皮细胞表达 E选择素和胞间粘附分子 1(ICAM- 1)的抑制作用。 方法 应用脂质体分别将 E选择素和 ICAM- 1ASODN转染内皮细胞 ,随后加入肿瘤坏死因子 α(TNF- α)诱导 ,并通过流式细胞技术测定内皮细胞膜 E选择素和 ICAM- 1蛋白的表达 ,RT- PCR半定量法测定 E选择素和ICAM- 1m RNA的量。 结果 裸 ASODN和脂质体 - ASODN均降低了内皮细胞 E选择素和 ICAM- 1分子及其m RNA的表达 ,脂质体 - ASODN尽管浓度低 ,作用更为明显。 结论
Objective\ To explore the inhibition of E\|selectin and intercellular adhesion molecule\|1(ICAM\|1) expression in endothelial cells by antisense oligodeoxynucleotides(ASODNs). Methods E\|\{selectin\} and ICAM\|1 ASODN liposome were introduced into the endothelial cells.\ E\|selectin and ICAM\|1 expression in the endothelial cell were induced by TNF\|α.\ The changes of E\|selectin and ICAM\|1 protein expression in endothelial cells were measured by flow cytometry, and E\|selectin and ICAM\|1 mRNA expression were examined by RT\|PCR. Results Both nude ASODN and liposome\|ASODN apparently brought down the protein and mRNA expression of E\|selectin and ICAM\|1, and the liposome\|ASODN worked better at lower dosage. Conclusion These data demonstrate that ASODN are capable of selectively inhibiting the expression of E\|selectin and ICAM\|1 in endothelial cells.\;
